(2004).&nbsp;<a href=\"http:\/\/molpharm.aspetjournals.org\/content\/65\/5\/1208\">Flavonoids are inhibitors of breast cancer resistance protein (ABCG2)-mediated transport.&nbsp;<\/a><em>Molecular pharmacology<\/em>, 65(5), 1208-1216.&nbsp;<\/p>\n\n\n\n<p>FONDO: La prote\u00edna de resistencia al c\u00e1ncer de mama (BCRP) es un nuevo transportador de uni\u00f3n a ATP, que se ha demostrado que confiere resistencia a m\u00faltiples f\u00e1rmacos (MDR) a varios agentes anticancer\u00edgenos importantes y desempe\u00f1a una funci\u00f3n importante en la regulaci\u00f3n de la disposici\u00f3n del f\u00e1rmaco. Los flavonoides son una clase de compuestos polifen\u00f3licos ampliamente presentes en alimentos y productos herbales. Las interacciones de los flavonoides con la glicoprote\u00edna P y la prote\u00edna 1 asociada a resistencia a m\u00faltiples f\u00e1rmacos han sido reportadas; sin embargo, su interacci\u00f3n con BCRP es desconocida.&nbsp;<\/p>\n\n\n\n<p>OBJETIVO: Nuestro objetivo fue evaluar los efectos de 20 flavonoides naturales sobre la acumulaci\u00f3n celular y la citotoxicidad de la mitoxantrona en l\u00edneas celulares humanas que sobreexpresan BCRP y BCRP.&nbsp;<\/p>\n\n\n\n<p>METODO:: En estos estudios se usaron c\u00e9lulas de c\u00e1ncer de mama humano BCRP que sobreexpresaban y BCRP-negativas (MCF-7) y c\u00e9lulas de carcinoma de pulm\u00f3n de c\u00e9lulas grandes (NCI-H460).&nbsp;<\/p>\n\n\n\n<p>RESULTADOS: Muchos de los flavonoides probados (50 \u03bcM) aumentaron la acumulaci\u00f3n de mitoxantrona en c\u00e9lulas que sobreexpresan BCRP, revirtiendo completamente la resistencia a mitoxantrona, sin efecto sobre las c\u00e9lulas BCRP negativas correspondientes, lo que indica que estos flavonoides son inhibidores de BCRP. Los efectos de estos flavonoides sobre la acumulaci\u00f3n celular y la citotoxicidad de la mitoxantrona dependieron de la concentraci\u00f3n de flavonoides, y se produjeron cambios significativos a concentraciones inferiores a 10 \u03bcM para la mayor\u00eda de los flavonoides. Crisina y biochanina A fueron los inhibidores de BCRP m\u00e1s potentes, produciendo aumentos significativos en la acumulaci\u00f3n de mitoxantrona a concentraciones de 0.5 o 1.0 \u03bcM y en citotoxicidad de mitoxantrona a una concentraci\u00f3n de 2.5 \u03bcM. Los gluc\u00f3sidos flavonoides no tuvieron efectos sobre el transporte de mitoxantrona mediado por BCRP.&nbsp;<\/p>\n\n\n\n<p>CONCLUSION: En conclusi\u00f3n, muchos flavonoides de origen natural pueden inhibir el flujo de salida mediado por BCRP y as\u00ed aumentar la acumulaci\u00f3n celular de sustratos de BCRP y restablecer la sensibilidad de las c\u00e9lulas de MDR. Algunos de estos flavonoides como la crisina y la biocanina A pueden ser efectivos a concentraciones muy bajas y potencialmente se podr\u00edan usar solos o en combinaci\u00f3n para revertir la MDR mediada por BCRP o como compuestos principales para el desarrollo de derivados \u00f3ptimos para la aplicaci\u00f3n cl\u00ednica. Desde el punto de vista de interacci\u00f3n con medicamentos, considerando el amplio consumo de grandes cantidades de flavonoides en alimentos que contienen flavonoides o productos a base de hierbas, las interacciones farmacocin\u00e9ticas de estos flavonoides con f\u00e1rmacos que son sustratos del BCRP, que producen alteraciones en la biodisponibilidad y eliminaci\u00f3n, podr\u00edan tener lugar despu\u00e9s coadministraci\u00f3n.\n\n<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Zhang, S., Yang, X., &amp; Morris, M.
