Sin embargo, el \nmecanismo molecular implicado en el efecto antimetast\u00e1sico de la \nbaicale\u00edna sigue siendo poco conocido. Este estudio tuvo como objetivo \nverificar los efectos inhibitorios de la baicale\u00edna sobre la met\u00e1stasis \nde las c\u00e9lulas de c\u00e1ncer de mama humano MDA-MB-231 tanto in vitro como \nin vivo, as\u00ed como investigar los mecanismos relacionados.<br><br>M\u00c9TODOS:\n El ensayo MTT se utiliz\u00f3 para examinar la inhibici\u00f3n de la baicale\u00edna \nen la proliferaci\u00f3n de c\u00e9lulas MDA-MB-231. El ensayo de curaci\u00f3n de \nheridas y el ensayo de invasi\u00f3n in vitro se llevaron a cabo para \ninvestigar los efectos de la baicale\u00edna en la migraci\u00f3n e invasi\u00f3n de \nc\u00e9lulas MDA-MB-231, respectivamente. Para explorar los efectos de la \nbaicale\u00edna sobre la met\u00e1stasis tumoral in vivo, se estableci\u00f3 el modelo \nde rat\u00f3n desnudo con xenoinjerto de c\u00e9lulas MDA-MB-231. Los animales se \ndividieron aleatoriamente en cuatro grupos (control, grupo de terapia y \ngrupo de prevenci\u00f3n de dosis bajas y altas, n = 6), y se trataron con \nbaicale\u00edna como se dise\u00f1\u00f3. Despu\u00e9s del sacrificio, se recogieron sus \npulmones e h\u00edgados para examinar la presencia de met\u00e1stasis. qRT-PCR y \nWestern blot se realizaron para estudiar los efectos de la baicale\u00edna en\n la expresi\u00f3n de SATB1, mol\u00e9culas relacionadas con EMT, y componentes de\n se\u00f1alizaci\u00f3n de Wnt \/ \u03b2-catenina de c\u00e9lulas MDA-MB-231, as\u00ed como del \ntejido metast\u00e1sico. Los efectos de la baicale\u00edna sobre la expresi\u00f3n de \nprote\u00ednas diana in vivo tambi\u00e9n se analizaron por inmunohistoqu\u00edmica.<\/p>\n\n\n\n<p>RESULTADOS:\n Nuestros resultados indicaron que la baicale\u00edna suprimi\u00f3 la \nproliferaci\u00f3n, la migraci\u00f3n y la invasi\u00f3n de las c\u00e9lulas MDA-MB-231 de \nuna manera dependiente del tiempo y la dosis. Seg\u00fan los ensayos \nrealizados en el modelo de rat\u00f3n desnudo con xenoinjerto, encontramos \nque la baicale\u00edna inhib\u00eda la met\u00e1stasis tumoral in vivo. Adem\u00e1s, la \nbaicale\u00edna disminuy\u00f3 significativamente la expresi\u00f3n de SATB1 en c\u00e9lulas\n MDA-MB-231. Suprimi\u00f3 la expresi\u00f3n de vimentina y CARACOL mientras \nmejoraba la expresi\u00f3n de E-cadherina. La baicale\u00edna tambi\u00e9n regulaba \nnegativamente la expresi\u00f3n de prote\u00ednas Wnt1 y \u03b2-catenina y el nivel de \ntranscripci\u00f3n de genes dirigidos a Wnt \/ \u03b2-catenina.<\/p>\n\n\n\n<p>CONCLUSI\u00d3N: \nNuestros resultados demuestran que la baicale\u00edna tiene el potencial de \nsuprimir la met\u00e1stasis del c\u00e1ncer de mama, posiblemente mediante la \ninhibici\u00f3n de la EMT, que puede atribuirse a la regulaci\u00f3n negativa \ntanto de SATB1 como de la v\u00eda Wnt \/ \u03b2-catenina.
