Sin embargo, el mecanismo anticancer\u00edgeno de la curcumina a\u00fan no se ha dilucidado por completo. La transici\u00f3n epitelial-mesenquimatosa (EMT) inducida por el factor de crecimiento transformante \u03b21 (TGF-\u03b21) participa en la promoci\u00f3n de la invasi\u00f3n tumoral y la met\u00e1stasis, y est\u00e1 estrechamente relacionada con la resistencia a los medicamentos de las c\u00e9lulas tumorales. La activaci\u00f3n anormal de la se\u00f1alizaci\u00f3n de Hedgehog tambi\u00e9n juega un papel importante en la tumorig\u00e9nesis y met\u00e1stasis. <\/p>\n<p>OBJETIVO \/ METODO: Para investigar si la curcumina puede revertir la EMT estimulada por TGF-\u03b21 de las c\u00e9lulas PANC-1 de c\u00e1ncer de p\u00e1ncreas, y su posible mecanismo, la l\u00ednea celular de c\u00e1ncer de p\u00e1ncreas PANC-1 fue estimulada con TGF-\u03b21 (5 ng \/ ml) durante 7 d\u00edas para inducir la formaci\u00f3n de EMT, y las c\u00e9lulas PANC-1 estimuladas con TGF-\u03b21 se trataron con diferentes concentraciones de curcumina (10, 20 y 30 \u00b5mol \/ ml) durante 48 h. <\/p>\n<p>MEDIDAS DE RESULTADOS: La tasa de inhibici\u00f3n del crecimiento de las c\u00e9lulas se midi\u00f3 mediante el ensayo MTT, la apoptosis se detect\u00f3 mediante citometr\u00eda de flujo, los niveles de expresi\u00f3n de Shh, GLI1, E-cadherina y vimentina se detectaron mediante an\u00e1lisis de transferencia Western, y la capacidad de invasi\u00f3n y migraci\u00f3n celular se examinaron mediante transwell ensayo de invasi\u00f3n celular y ensayo de cicatrizaci\u00f3n de heridas. <\/p>\n<p>RESULTADOS: Despu\u00e9s de la estimulaci\u00f3n con TGF-\u03b21, los niveles de expresi\u00f3n de Shh, GLI1 y vimentina en el grupo estimulado por TGF-\u03b21 aumentaron significativamente, en comparaci\u00f3n con los del grupo control (P &lt;0.01, respectivamente). Los niveles de expresi\u00f3n de E-cadherina en el grupo estimulado con TGF-\u03b21 disminuyeron significativamente, en comparaci\u00f3n con los del grupo control (P &lt;0.01). Las c\u00e9lulas PANC-1 estimuladas con TGF-\u03b21 se trataron con curcumina y los resultados mostraron que la curcumina inhibi\u00f3 significativamente la proliferaci\u00f3n de c\u00e9lulas PANC-1 estimuladas con TGF-\u03b21 y la apoptosis inducida, en comparaci\u00f3n con otros grupos (P &lt;0.01), y los niveles de expresi\u00f3n de Shh, GLI1 y vimentina en el grupo tratado con curcumina disminuyeron significativamente en comparaci\u00f3n con los del grupo control (P &lt;0.01, respectivamente). El nivel de expresi\u00f3n de E-cadherina en el grupo tratado con curcumina aument\u00f3 significativamente en comparaci\u00f3n con el del grupo control (P &lt;0.01). La invasi\u00f3n celular en el grupo tratado con curcumina (30 \u00b5mol \/ ml) disminuy\u00f3 significativamente en comparaci\u00f3n con la del grupo control (P &lt;0.01). Las heridas por ara\u00f1azos en el grupo tratado con curcumina se curaron m\u00e1s lentamente en comparaci\u00f3n con las del grupo control (P &lt;0.01). La curcumina inhibi\u00f3 significativamente la invasi\u00f3n y migraci\u00f3n de c\u00e9lulas PANC-1 estimuladas con TGF-\u03b21. <\/p>\n<p>CONCLUSION: Estos resultados indican que la curcumina puede inhibir la proliferaci\u00f3n de c\u00e9lulas PANC-1 estimuladas por TGF-\u03b21, puede inducir apoptosis y revertir la EMT.
