Sin embargo, los efectos potenciales de la capsaicina en las c\u00e9lulas de osteosarcoma y los mecanismos subyacentes no se entienden completamente.\u00a0<\/p>\n<p>En el estudio actual, observamos que la inhibici\u00f3n del crecimiento inducida por capsaicina y la apoptosis en c\u00e9lulas de osteosarcoma cultivadas (U2OS y MG63), que se asociaron con una activaci\u00f3n significativa de la prote\u00edna quinasa activada por AMP (AMPK). La inhibici\u00f3n de AMPK por la interferencia del compuesto C o ARN suprimi\u00f3 la citotoxicidad inducida por capsaicina, mientras que los activadores de AMPK (AICAR y A769662) promovieron la muerte celular por osteosarcoma.\u00a0<\/p>\n<p>Para el estudio del mecanismo, encontramos que se requer\u00eda la activaci\u00f3n de AMPK para la inhibici\u00f3n de mTORC1 inducida por capsaicina (complejo mTOR 1), la regulaci\u00f3n negativa del linfoma 2 de c\u00e9lulas B (Bcl-2) y la regulaci\u00f3n positiva de Bax en c\u00e9lulas MG63. La administraci\u00f3n de capsaicina indujo la activaci\u00f3n de p53, translocaci\u00f3n mitocondrial y asociaci\u00f3n asesina Bcl-2, tales efectos depend\u00edan de la activaci\u00f3n de AMPK.\u00a0<\/p>\n<p>Curiosamente, observamos una activaci\u00f3n significativa de cinasas pro-apopt\u00f3ticas NH2-terminal cinasas por capsaicina en c\u00e9lulas MG63, que parec\u00eda ser independiente de AMPK.\u00a0<\/p>\n<p>En conclusi\u00f3n, la capsaicina pose\u00eda una fuerte eficacia contra las c\u00e9lulas de osteosarcoma humano. Los estudios moleculares revelaron que la capsaicina activ\u00f3 las se\u00f1ales dependientes de AMPK e independientes de AMPK para mediar en la apoptosis celular.
