(2006).\u00a0<a href=\"https:\/\/www.karger.com\/Article\/Abstract\/93790\">Anti-inflammatory activity of thymol: inhibitory effect on the release of human neutrophil elastase<\/a>.\u00a0<i>Pharmacology<\/i>, 77(3), 130-136.<\/p>\n<p>La elastasa, una serina proteinasa liberada por neutr\u00f3filos humanos activados, puede degradar una amplia variedad de biomacromol\u00e9culas, incluida la elastina, y se considera un marcador de enfermedades inflamatorias.\u00a0<\/p>\n<p>Como la estrategia l\u00f3gica para proteger el tejido es inhibir la actividad excesiva de elastasa, las investigaciones experimentales y cl\u00ednicas se han concentrado en tratar de encontrar inhibidores de elastasa eficientes.\u00a0<\/p>\n<p>Como el timol, uno de los componentes principales del aceite de tomillo con estructura fen\u00f3lica, ha sido acreditado con una serie de propiedades farmacol\u00f3gicas, que incluyen efectos antimicrobianos y antioxidantes, el objetivo de este estudio fue explorar si tambi\u00e9n puede interferir con la liberaci\u00f3n de elastasa por neutr\u00f3filos humanos estimulados con el p\u00e9ptido quimiot\u00e1ctico sint\u00e9tico N-formil-metionil-leucil-fenilalanina (fMLP).\u00a0<\/p>\n<p>Despu\u00e9s de incubar los neutr\u00f3filos con cantidades crecientes de timol (2.5, 5, 10, 20 \u00b5g \/ ml), se inici\u00f3 la liberaci\u00f3n de elastasa por fMLP y se midi\u00f3 usando MeO-Suc-Ala-Ala-Pro-Val-MCA.\u00a0<\/p>\n<p>Los resultados mostraron que el timol inhibi\u00f3 la liberaci\u00f3n de elastasa inducida por fMLP de una manera dependiente de la concentraci\u00f3n, siendo los efectos de 10 y 20 \u00b5g \/ ml estad\u00edsticamente significativos. El comportamiento de la movilizaci\u00f3n de calcio citos\u00f3lico revelado por fura-2 se parec\u00eda mucho al de la elastasa, lo que sugiere que pueden estar relacionados. La naturaleza hidrof\u00f3bica del timol significa que puede acercarse a las prote\u00ednas del canal i\u00f3nico a trav\u00e9s de la fase lip\u00eddica de la membrana, alterar el entorno local de los canales de calcio e inhibir as\u00ed la entrada capacitiva de calcio.\u00a0<\/p>\n<p>En resumen, el timol inactiva la maquinaria de los canales de calcio, lo que desencadena una reducci\u00f3n correspondiente en la elastasa. La actividad antibacteriana y antimic\u00f3tica del timol ya es bien conocida, pero nuestros hallazgos de que inhibe la elastasa ampl\u00edan nuestro conocimiento de la actividad antiinflamatoria de esta mol\u00e9cula interesante que ya est\u00e1 acreditada con actividad antioxidante.
