La evidencia existente ha revelado que el bloqueo de la gluc\u00f3lisis produce la apoptosis de las c\u00e9lulas cancerosas. Tanshinone IIA (Tan IIA) es una naftoquinona diterpenoide que se encuentra en la medicina tradicional china, Danshen (Salvia sp.). Tan IIA exhibe actividad potencial contra el c\u00e1ncer. Sin embargo, se desconoce su efecto sobre la viabilidad celular de las c\u00e9lulas de c\u00e1ncer cervical humano y su mecanismo.\u00a0<\/p>\n<p>OBJETIVO: El objetivo del presente estudio fue determinar el efecto de Tan IIA sobre la proliferaci\u00f3n y el metabolismo de la glucosa en las c\u00e9lulas de c\u00e1ncer de cuello uterino.\u00a0<\/p>\n<p>MATERIALES \/ MEDIDAS \/ M\u00c9TODO: La viabilidad celular se midi\u00f3 mediante el ensayo MTT, la apoptosis se determin\u00f3 utilizando citometr\u00eda de flujo y la absorci\u00f3n de glucosa, la producci\u00f3n de lactato y el contenido de adenosina trifosfato se midieron para evaluar el metabolismo de la glucosa. La expresi\u00f3n de prote\u00ednas asociadas a la apoptosis se detect\u00f3 mediante transferencia Western y se evalu\u00f3 la actividad antitumoral de Tan IIA in vivo en ratones portadores de carcinoma cervical.\u00a0<\/p>\n<p>RESULTADOS: Los resultados revelaron que el tratamiento con Tan IIA result\u00f3 en una reducci\u00f3n considerable en la viabilidad de las c\u00e9lulas SiHa. Tan IIA disminuy\u00f3 la expresi\u00f3n de los oncogenes E6 y E7 del VPH, indujo la apoptosis y tambi\u00e9n disminuy\u00f3 la gluc\u00f3lisis al suprimir la actividad del AKT \/ mTOR intracelular y HIF-1\u03b1. In vivo, el tratamiento con Tan IIA dio como resultado una reducci\u00f3n del 72,7% en el volumen del tumor.\u00a0<\/p>\n<p>CONCLUSI\u00d3N: El presente estudio destaca el valor terap\u00e9utico potencial de Tan IIA, que funciona al inducir apoptosis y puede estar asociado con la inhibici\u00f3n de la gluc\u00f3lisis.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Liu, Z., Zhu, W., Kong, X., Chen, X., Sun, X., Zhang, W., &amp; Zhang, R.
