(2016).\u00a0<a href=\"https:\/\/link.springer.com\/article\/10.1007\/s13277-015-4602-9\">The interplay between autophagy and apoptosis induced by tanshinone IIA in prostate cancer cells<\/a>.\u00a0<i>Tumor Biology<\/i>, 37(6), 7667-7674.<\/p>\n<p>CONTEXTO: La tanshinona IIA (T2A), un derivado de fenantrenoquinona y tambi\u00e9n el principal ingrediente activo de Danshen, ha recibido mucha atenci\u00f3n como una prometedora terapia contra el c\u00e1ncer por sus posibles actividades contra el c\u00e1ncer.\u00a0<\/p>\n<p>OBJETIVO \/ M\u00c9TODO \/ MEDIDAS: En este estudio, se observ\u00f3 la apoptosis y la autofagia de las c\u00e9lulas PC-3 de c\u00e1ncer de pr\u00f3stata humano despu\u00e9s del tratamiento con T2A 5 \u03bcM, as\u00ed como su relevancia. La apoptosis dependiente de mitocondrias se detect\u00f3 en primer lugar mediante observaci\u00f3n morfol\u00f3gica y an\u00e1lisis bioqu\u00edmico. Mientras tanto, T2A 5 \u03bcM desencaden\u00f3 con \u00e9xito la autofagia de las c\u00e9lulas PC-3, indicada por una mayor expresi\u00f3n de Beclin1 y LC3 II.\u00a0<\/p>\n<p>RESULTADOS: Se realizaron experimentos de validaci\u00f3n para consolidar a\u00fan m\u00e1s la contribuci\u00f3n de T2A a la autofagia: el pretratamiento con inhibidor de la autofagia 3-metiladenina (3-MA) \u200b\u200bproporcion\u00f3 protecci\u00f3n contra la autofagia y la apoptosis inducida por T2A mejorada. Adem\u00e1s, el supresor de apoptosis z-VAD-fmk no pudo facilitar la formaci\u00f3n de vacuolas autof\u00e1gicas, lo que tambi\u00e9n demostr\u00f3 que la autofagia inducida por T2A es independiente de la apoptosis. Adem\u00e1s, el eliminador de especies reactivas de ox\u00edgeno (ROS) N-acetil-La 1 -ciste\u00edna (NAC) inhibi\u00f3 eficazmente la expresi\u00f3n de Beclin1, LC3-II y caspasa-3 escindida, lo que indicaba apoptosis y autofagia con dependencia de la producci\u00f3n intracelular de ROS.\u00a0<\/p>\n<p>CONCLUSIONES: Tomados en conjunto, estos resultados demostraron que la autofagia es el mecanismo citoprotector en este sistema experimental, y los ROS resultantes del tratamiento con T2A desempe\u00f1aron un papel cr\u00edtico en la apoptosis y el inicio de la autofagia.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Li, C., Han, X., Zhang, H., Wu, J., &amp; Li, B.
