El \u00e9xito de esta actividad sint\u00e9tica depende de la activaci\u00f3n de las v\u00edas para generar potencia reductora (NADPH) y restaurar el oxaloacetato para la funci\u00f3n continua del ciclo TCA (anaplerosis). Sorprendentemente, ambas necesidades fueron satisfechas por una alta tasa de metabolismo de la glutamina.\u00a0<\/p>\n<p>Primero, la conversi\u00f3n de glutamina en lactato (glutamin\u00f3lisis) fue lo suficientemente r\u00e1pida como para producir suficiente NADPH para soportar la s\u00edntesis de \u00e1cidos grasos.\u00a0<\/p>\n<p>En segundo lugar, a pesar del importante metabolismo mitocondrial del piruvato, se suprimi\u00f3 la carboxilaci\u00f3n del piruvato, y el oxaloacetato anapler\u00f3tico se deriv\u00f3 de la glutamina. El catabolismo de la glutamina estuvo acompa\u00f1ado por la secreci\u00f3n de alanina y amon\u00edaco, de modo que la mayor\u00eda de los grupos amino de la glutamina se perdieron de la c\u00e9lula en lugar de incorporarse a otras mol\u00e9culas.\u00a0<\/p>\n<p>Estos datos demuestran que las c\u00e9lulas transformadas exhiben una alta tasa de consumo de glutamina que no puede explicarse por la demanda de nitr\u00f3geno impuesta por la s\u00edntesis de nucle\u00f3tidos o el mantenimiento de grupos de amino\u00e1cidos no esenciales.
