(2006).\u00a0<a href=\"https:\/\/iovs.arvojournals.org\/article.aspx?articleid=2124858\">Flavonoids protect human retinal pigment epithelial cells from oxidative-stress\u2013induced death<\/a>.\u00a0<i>Investigative ophthalmology &amp; visual science,<\/i>\u00a047(7), 3164-3177.<\/p>\n<p>PROP\u00d3SITO: Determinar si los flavonoides diet\u00e9ticos y sint\u00e9ticos espec\u00edficos pueden proteger las c\u00e9lulas epiteliales del pigmento retiniano humano (RPE) de la muerte inducida por el estr\u00e9s oxidativo.<\/p>\n<p>M\u00c9TODOS: La eficacia y la potencia se determinaron de una variedad de flavonoides en la dieta y sint\u00e9ticos sobre la supervivencia de c\u00e9lulas ARPE-19 humanos y c\u00e9lulas de RPE primarias humanas tratadas con per\u00f3xido de hidr\u00f3geno (H 2 O 2 ) o t -butil hidroper\u00f3xido ( t- BOOH). Determinamos las concentraciones efectivas (CE 50 s) y las toxicidades (LD 50s) de los flavonoides despu\u00e9s de 24 horas, utilizando el ensayo MTT. La eficacia de las vitaminas E y C en la supervivencia de las c\u00e9lulas RPE se compar\u00f3 en condiciones id\u00e9nticas. La capacidad de los flavonoides espec\u00edficos para proteger las c\u00e9lulas RPE de la muerte celular se determin\u00f3 en varios intervalos de tiempo despu\u00e9s de que las c\u00e9lulas fueron expuestas al estr\u00e9s oxidativo. La capacidad de los flavonoides para bloquear la acumulaci\u00f3n de especies de ox\u00edgeno reactivo intracelular se examin\u00f3 con fluorescencia de diclorofluoresce\u00edna (DCF). Finalmente, la capacidad de los flavonoides para inducir enzimas desintoxicantes de fase 2 se prob\u00f3 mediante an\u00e1lisis de inmunotransferencia para el factor de transcripci\u00f3n Nrf2 y el producto g\u00e9nico de fase 2 heme-oxigenasa 1.<\/p>\n<p>RESULTADOS: Los flavonoides espec\u00edficos protegieron a las c\u00e9lulas RPE humanas de la muerte inducida por el estr\u00e9s oxidativo con una eficacia entre 80% y 100% y potencias en el rango alto nanomolar y bajo micromolar. Las toxicidades de la mayor\u00eda de los flavonoides efectivos fueron bajas. Los flavonoides efectivos incluyeron los flavonoides diet\u00e9ticos fisetina, luteolina, quercetina, eriodictyol, baicale\u00edna, galangin y EGCG, y los flavonoides sint\u00e9ticos, 3,6-dihidroxiflavonol y 3,7 dihidroxiflavonol. Varios flavonoides pueden proteger las c\u00e9lulas RPE incluso cuando se agregan despu\u00e9s de que las c\u00e9lulas han estado expuestas al estr\u00e9s oxidativo. Los flavonoides actuaron a trav\u00e9s de una ruta intracelular para bloquear la acumulaci\u00f3n de especies reactivas de ox\u00edgeno. Muchos de estos flavonoides indujeron la expresi\u00f3n de Nrf2 y el producto g\u00e9nico de la fase 2 hemooxigenasa 1 en c\u00e9lulas RPE humanas.<\/p>\n<p>CONCLUSIONES: Los resultados identifican un grupo selecto de flavonoides que protegen las c\u00e9lulas RPE de la muerte inducida por estr\u00e9s oxidativo con un alto grado de potencia y baja toxicidad. Muchos de estos flavonoides tambi\u00e9n inducen la expresi\u00f3n de prote\u00ednas de desintoxicaci\u00f3n de fase 2 que podr\u00edan funcionar para proporcionar protecci\u00f3n adicional contra el estr\u00e9s oxidativo.
