Adem\u00e1s, los niveles intracelulares de antioxidantes y las enzimas depuradoras de radicales libres se alteran gradualmente.\u00a0<\/p>\n<p>Estos dos factores de combinaci\u00f3n conducen a un aumento dependiente de la edad en la fracci\u00f3n de ROS y radicales libres que pueden escapar del mecanismo de defensa y causar da\u00f1o oxidativo a varias biomol\u00e9culas en las c\u00e9lulas de los tejidos.\u00a0<\/p>\n<p>Un creciente cuerpo de evidencia ha establecido que los niveles de ROS y el da\u00f1o oxidativo a l\u00edpidos, prote\u00ednas y \u00e1cidos nucleicos aumentan significativamente con la edad en tejidos animales y humanos. El ADN mitocondrial (ADNmt), aunque no est\u00e1 protegido por histonas o prote\u00ednas de uni\u00f3n al ADN, es susceptible al da\u00f1o oxidativo por los niveles cada vez mayores de ROS y radicales libres en la matriz mitocondrial.\u00a0<\/p>\n<p>En los \u00faltimos a\u00f1os, se ha encontrado que la modificaci\u00f3n oxidativa (formaci\u00f3n de 8-hidroxi-2&#8242;-desoxiguanosina) y la supresi\u00f3n a gran escala y la mutaci\u00f3n puntual del mtDNA aumentan exponencialmente con la edad en diversos tejidos humanos. Las enzimas respiratorias que contienen las subunidades proteicas defectuosas codificadas por mtDNA mutante exhiben inevitablemente una funci\u00f3n respiratoria deteriorada y de ese modo aumentan la fuga de electrones y la producci\u00f3n de ROS, que a su vez eleva el estr\u00e9s oxidativo y el da\u00f1o oxidativo de las mitocondrias.\u00a0<\/p>\n<p>Este c\u00edrculo vicioso opera en diferentes c\u00e9lulas de tejidos a diferentes velocidades y por lo tanto conduce a la acumulaci\u00f3n diferencial de la mutaci\u00f3n y el da\u00f1o oxidativo del ADN mitocondrial en el envejecimiento humano.
