(2006).\u00a0<a href=\"https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/S0006295206003704\">Caffeine metabolites are inhibitors of the nuclear enzyme poly (ADP-ribose) polymerase-1 at physiological concentrations.<\/a>\u00a0<i>Biochemical pharmacology<\/i>, 72(7), 902-910.<\/p>\n<p>La actividad de la enzima nuclear poli (ADP-ribosa) polimerasa-1 (EC2.4.2.30), altamente activada por roturas de la cadena de ADN, est\u00e1 asociada con la fisiopatolog\u00eda de enfermedades inflamatorias agudas y cr\u00f3nicas. La sobreactivaci\u00f3n de PARP-1 y la extensa renovaci\u00f3n posterior de su sustrato NAD + ponen una gran demanda en la producci\u00f3n mitocondrial de ATP. Adem\u00e1s, debido a su papel informado en la producci\u00f3n de citocinas proinflamatorias mediadas por NF-\u03baB y AP-1, PARP-1 se considera un objetivo interesante en el tratamiento de estas enfermedades.<\/p>\n<p>En este estudio, se prob\u00f3 la capacidad inhibidora de PARP-1 de la cafe\u00edna y varios metabolitos, as\u00ed como otras (metil) xantinas utilizando un ensayo ELISA con PARP-1 humano purificado. La cafe\u00edna en s\u00ed misma mostr\u00f3 solo una d\u00e9bil actividad inhibidora de PARP-1, mientras que los metabolitos de cafe\u00edna 1,7-dimetilxantina, 3-metilxantina y 1-metilxantina, as\u00ed como teobromina y teofilina mostraron una actividad inhibidora de PARP-1 significativa. La evaluaci\u00f3n adicional de estos compuestos en H 2 O 2 tratado con A549 de pulm\u00f3n epitelial y RF24 vascular endotelial c\u00e9lulas revelaron que la disminuci\u00f3n de NAD + -los niveles, as\u00ed como la formaci\u00f3n de la poli (ADP-ribosa) pol\u00edmero fue significativamente impedido por el importante cafe\u00edna metabolito 1,7-dimetilxantina. Adem\u00e1s, H 2 O 2inducida por la necrosis podr\u00eda prevenirse con una dosis alta de 1,7-dimetilxantina. Finalmente, los efectos antioxidantes de las metilxantinas podr\u00edan descartarse con la VSG y la medici\u00f3n del TEAC.<\/p>\n<p>En conclusi\u00f3n, los metabolitos de cafe\u00edna son inhibidores de PARP-1 y el metabolito principal de cafe\u00edna 1,7-dimetilxantina tiene una actividad inhibidora significativa de PARP-1 en c\u00e9lulas epiteliales y endoteliales cultivadas a concentraciones fisiol\u00f3gicas.
