La hiperglucemia ha sido implicada como un importante contribuyente a varias complicaciones de la diabetes. Los niveles altos de glucosa inducen la liberaci\u00f3n de citocinas proinflamatorias. La luteolina es una flavona aislada del apio, el pimiento verde, la hoja de perilla y el t\u00e9 de manzanilla. Se ha informado que la luteolina posee propiedades antimutag\u00e9nicas, antitumorales, antioxidantes y antiinflamatorias.\u00a0<\/p>\n<p>OBJETIVO: En este estudio, investigamos los efectos de la luteolina en la secreci\u00f3n de citocinas proinflamatorias y su regulaci\u00f3n epigen\u00e9tica subyacente en monocitos humanos con alto contenido de glucosa.\u00a0<\/p>\n<p>M\u00c9TODO \/ DISE\u00d1O: Las c\u00e9lulas monoc\u00edticas humanas (THP-1) se cultivaron en condiciones controladas (manitol 14,5 mM), normogluc\u00e9mico (NG, glucosa 5,5 mM) o hipergluc\u00e9mico (HG, glucosa 20 mM), en ausencia o presencia de luteolina. Se a\u00f1adi\u00f3 luteolina (3-10 \u03bcM) durante 48 h.\u00a0<\/p>\n<p>RESULTADOS: Mientras que las condiciones hipergluc\u00e9micas indujeron significativamente la acetilaci\u00f3n de histonas, la activaci\u00f3n de NF-\u03baB y la liberaci\u00f3n de citocinas proinflamatorias (IL-6 y TNF-\u03b1) de las c\u00e9lulas THP-1, la luteolina suprimi\u00f3 la actividad de NF-\u03baB y la liberaci\u00f3n de citocinas. La luteolina tambi\u00e9n redujo significativamente la expresi\u00f3n g\u00e9nica de la prote\u00edna de uni\u00f3n a CREB \/ p300 (CBP \/ p300), as\u00ed como los niveles de acetilaci\u00f3n y actividad de la histona acetiltransferasa (HAT) de la prote\u00edna CBP \/ p300, que es un coactivador NF-\u03baB conocido.\u00a0<\/p>\n<p>CONCLUSIONES: Estos resultados sugieren que la luteolina inhibe la producci\u00f3n de citocinas inducida por HG en monocitos, a trav\u00e9s de cambios epigen\u00e9ticos que involucran NF-\u03baB.
