Recientemente se ha propuesto que la protecci\u00f3n de la funci\u00f3n mitocondrial (que est\u00e1 alterada en varias enfermedades como el Alzheimer, el Parkinson, la obesidad y la diabetes) por estos compuestos, puede ser importante para explicar los efectos beneficiosos de los polifenoles en la salud.\u00a0<\/p>\n<p>OBJETIVO \/ M\u00c9TODO: El objetivo de este estudio fue evaluar los efectos protectores de los polifenoles diet\u00e9ticos de quercetina, rutina, resveratrol y galato de epigalocatequina contra las alteraciones de la funci\u00f3n mitocondrial inducida por la indometacina (INDO) en las c\u00e9lulas epiteliales intestinales de Caco-2, y abordar el mecanismo involucrado en tales efecto nocivo por INDO, que genera estr\u00e9s oxidativo. La concentraci\u00f3n de INDO disminuye de manera dependiente los niveles de ATP celular y el potencial de membrana mitocondrial en las c\u00e9lulas Caco-2 despu\u00e9s de 20 minutos de incubaci\u00f3n. INDO tambi\u00e9n inhibe la actividad del complejo mitocondrial I y provoca la acumulaci\u00f3n de NADH; que conduce a la sobreproducci\u00f3n de O mitocondrial (2) () (-), ya que es evitado por el piruvato.\u00a0<\/p>\n<p>RESULTADOS: La quercetina (0,01 mg \/ ml), el resveratrol (0,1 mg \/ ml) y la rutina (1 mg \/ ml) protegieron las c\u00e9lulas Caco-2 contra la disfunci\u00f3n mitocondrial inducida por INDO, mientras que no se observ\u00f3 protecci\u00f3n con galato de epigalocatequina. La quercetina fue la m\u00e1s eficiente en la protecci\u00f3n contra la disfunci\u00f3n mitocondrial; Esto podr\u00eda deberse a su capacidad para ingresar a las c\u00e9lulas y acumularse en las mitocondrias. Adem\u00e1s, su similitud estructural con la rotenona podr\u00eda favorecer su uni\u00f3n al sitio ubiquinona del complejo I, protegi\u00e9ndolo de inhibidores como INDO o rotenona.\u00a0<\/p>\n<p>CONCLUSI\u00d3N: Estos hallazgos sugieren un posible nuevo papel protector para los polifenoles de la dieta para las mitocondrias, complementario de su propiedad antioxidante.
