SHP2, SOCS3 y PIAS3 son conocidos como los reguladores negativos de la se\u00f1alizaci\u00f3n de STAT3, mientras que su relevancia para la activaci\u00f3n frecuente de STAT3 en meduloblastomas sigue siendo desconocida.\u00a0<\/p>\n<p>M\u00c9TODOS: Se construyeron microarrays de tejido con 17 tejidos cerebrales no cancerosos que rodean el tumor y 61 casos de meduloblastomas cl\u00e1sicos, 44 meduloblastomas de c\u00e9lulas grandes y 15 meduloblastomas nodulares, que se usaron para el perfil inmunohistoqu\u00edmico de patrones de expresi\u00f3n STAT3, SHP2, SOCS3 y PIAS3 y las frecuencias de translocaci\u00f3n nuclear STAT3. Se cultivaron tres l\u00edneas celulares de meduloblastoma humano (Daoy, UW228-2 y UW228-3) con y sin suplemento de resveratrol 100 \u03bcM. Las influencias del resveratrol en SHP2, La expresi\u00f3n de SOCS3 y PIAS3 y la ca\u00edda de SOCS3 en la activaci\u00f3n de STAT3 se analizaron utilizando m\u00faltiples enfoques experimentales.\u00a0<\/p>\n<p>RESULTADO: Los niveles de SHP2, SOCS3 y PIAS3 se reducen en meduloblastomas in vivo e in vitro, de los cuales la regulaci\u00f3n por disminuci\u00f3n de PIAS3 se correlaciona m\u00e1s inversamente con la activaci\u00f3n de STAT3. En las c\u00e9lulas de meduloblastoma con supresi\u00f3n del resveratrol con regulaci\u00f3n negativa de STAT3 y disminuci\u00f3n de la incidencia de la translocaci\u00f3n nuclear de STAT3, PIAS3 est\u00e1 regulado al alza, el nivel de SHP2 permanece sin cambios y la regulaci\u00f3n a la baja de SOCS3. Las prote\u00ednas SOCS3 se acumulan en los extremos distales de los procesos de tipo ax\u00f3n de las c\u00e9lulas de meduloblastoma diferenciadas con resveratrol. La eliminaci\u00f3n de la expresi\u00f3n de SOCS3 por siRNA no influye en la proliferaci\u00f3n celular ni en la activaci\u00f3n de STAT3 o en la sensibilidad al resveratrol, pero inhibe la formaci\u00f3n de procesos similares a axones inducidos por el resveratrol.\u00a0<\/p>\n<p>CONCLUSI\u00d3N: Nuestros resultados sugieren que (1) la reducci\u00f3n general de SHP2, SOCS3 y PIAS3 en tejidos de meduloblastoma y l\u00edneas celulares; (2) la relevancia m\u00e1s inversa de la expresi\u00f3n de PIAS3 con la activaci\u00f3n de STAT3; (3) los valores pron\u00f3sticos favorables de PIAS3 para meduloblastomas y (4) la participaci\u00f3n de SOCS3 en la regeneraci\u00f3n de ax\u00f3n de c\u00e9lulas de meduloblastoma promovida por resveratrol.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Li, C., Li, H., Zhang, P., Yu, L.
