(2007).\u00a0<a href=\"https:\/\/www.pnas.org\/content\/pnas\/104\/13\/5419.full.pdf\">SHP-2 tyrosine phosphatase inhibits p73-dependent apoptosis and expression of a subset of p53 target genes induced by EGCG<\/a>.<i>\u00a0Proceedings of the National Academy of Sciences<\/i>, 104(13), 5419-5424.<\/p>\n<p>El polifenol del t\u00e9 verde, epigalocatequina-3-galato (EGCG) regula diferencialmente el crecimiento celular de las c\u00e9lulas cancerosas de una manera dependiente de p53 a trav\u00e9s de la apoptosis y \/ o la detenci\u00f3n del ciclo celular.\u00a0<\/p>\n<p>En un esfuerzo por dilucidar a\u00fan m\u00e1s el mecanismo de regulaci\u00f3n del crecimiento diferencial por EGCG, hemos investigado el papel de la tirosina fosfatasa, SHP-2. Al comparar las respuestas de los fibroblastos embrionarios de rat\u00f3n (MEF), que expresan WT o SHP-2 funcionalmente inactivo \/ truncado, encontramos que la inactivaci\u00f3n de SHP-2 sensibiliza notablemente las c\u00e9lulas a la muerte mediada por EGCG.\u00a0<\/p>\n<p>Los MEF que carecen de SHP-2 funcional sufren una apoptosis masiva tras el tratamiento con EGCG. Al comparar los perfiles de expresi\u00f3n g\u00e9nica, hemos identificado un conjunto de objetivos transcripcionales de p53 que se modulan diferencialmente en c\u00e9lulas que sufren apoptosis. Los an\u00e1lisis de Western blot y PCR en tiempo real de un grupo selecto de genes confirman adem\u00e1s que la expresi\u00f3n depende de SHP-2.\u00a0<\/p>\n<p>Se hicieron observaciones similares en MEF que carecen de p53, lo que confirma que la expresi\u00f3n de estos \u00abgenes diana de p53\u00bb es independiente de p53. Adem\u00e1s, el tratamiento con EGCG indujo la expresi\u00f3n de ARNm y prote\u00edna p73 en ambos tipos de c\u00e9lulas, pero no en p63. La inactivaci\u00f3n de p73 en c\u00e9lulas que expresan SHP-2 no funcional inhibi\u00f3 notablemente la apoptosis y la expresi\u00f3n del gen diana p53.\u00a0<\/p>\n<p>Aunque la fosforilaci\u00f3n de JNK est\u00e1 regulada diferencialmente por SHP2, se descubri\u00f3 que era prescindible para la apoptosis inducida por EGCG y la expresi\u00f3n del gen diana p53.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Amin, A.
