Las especies de ox\u00edgeno reactivo aumentado (ROS) y el alto flujo glucol\u00edtico son caracter\u00edsticas comunes de las c\u00e9lulas malignas.\u00a0<\/p>\n<p>OBJETIVO: Por lo tanto, evaluamos el efecto del resveratrol sobre el metabolismo de la glucosa en las c\u00e9lulas cancerosas e investigamos el papel de ROS en la respuesta.<\/p>\n<p>M\u00c9TODOS: Las c\u00e9lulas cancerosas se midieron para el contenido celular y (18) absorci\u00f3n de F-FDG. Se realizaron ensayos para la producci\u00f3n de lactato; actividad hexoquinasa y ROS intracelular; e inmunotransferencia para el factor inducible por hipoxia-1\u03b1 (HIF-1\u03b1), Akt, blanco mam\u00edfero de rapamicina y transportador de glucosa tipo 1 (Glut-1). Los estudios en animales se realizaron con im\u00e1genes PET de animales peque\u00f1os de ratones con tumor de carcinoma de pulm\u00f3n de Lewis.<\/p>\n<p>RESULTADOS: El resveratrol disminuy\u00f3 levemente el contenido celular y la absorci\u00f3n de F-FDG suprimida (18) de manera m\u00e1s pronunciada en el carcinoma de pulm\u00f3n de Lewis, el colon HT-29 y las c\u00e9lulas de c\u00e1ncer de mama T47D. Por lo tanto, (18) la absorci\u00f3n de F-FDG normalizada al contenido celular se redujo a menos de la mitad de los controles por la exposici\u00f3n de 24 h al resveratrol. Esta reducci\u00f3n se atribuy\u00f3 al flujo glucol\u00edtico reducido y a la expresi\u00f3n de Glut-1. El resveratrol tambi\u00e9n disminuy\u00f3 la ROS intracelular en patrones que eran muy parecidos a la absorci\u00f3n de F-FDG (18). La eliminaci\u00f3n de ROS con N-acetil ciste\u00edna, pero no la inhibici\u00f3n de la nicotinamida adenina dinucle\u00f3tido fosfato oxidasa, fue suficiente para suprimir (18) la absorci\u00f3n de F-FDG. Por el contrario, los inductores ROS invirtieron efectivamente la respuesta metab\u00f3lica del resveratrol. La prote\u00edna HIF-1\u03b1 se redujo notablemente por el resveratrol, e inhibir la expresi\u00f3n de HIF-1\u03b1 con cicloheximida o peque\u00f1os ARN interferentes espec\u00edficos suprimieron (18) la absorci\u00f3n de F-FDG. El inhibidor proteos\u00f3mico MG132 restaur\u00f3 parcialmente el nivel de HIF-1\u03b1 y (18) la absorci\u00f3n de F-FDG en las c\u00e9lulas tratadas con resveratrol. El resveratrol tambi\u00e9n inhibi\u00f3 la activaci\u00f3n de Akt; Adem\u00e1s, los inhibidores y los peque\u00f1os ARN interferentes contra la fosfoinositida 3-quinasa disminuyeron (18) la absorci\u00f3n de F-FDG. Finalmente, los resultados de PET de animales peque\u00f1os mostraron tratamiento con resveratrol para suprimir la absorci\u00f3n de F-FDG del tumor (18) in vivo.<\/p>\n<p>CONCLUSI\u00d3N: El resveratrol suprime la captaci\u00f3n de F-FDG de las c\u00e9lulas cancerosas (18) y el metabolismo glucol\u00edtico de una manera que depende de la capacidad del resveratrol para inhibir la ROS intracelular, que regula a la baja la acumulaci\u00f3n de HIF-1\u03b1.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Jung, K.
