International journal of oncology, 20(3), 631-636.<\/p>\n<p>FONDO: La relaci\u00f3n entre el consumo de vegetales cruc\u00edferos y la reducci\u00f3n de la aparici\u00f3n de c\u00e1ncer de pr\u00f3stata ha sido documentada, aunque se desconocen los fitoqu\u00edmicos responsables.<\/p>\n<p>OBJETIVO: Se investigaron los efectos del sulforafano (SFN) que ocurre como el precursor del glucosinolato en el br\u00f3coli y otros vegetales cruc\u00edferos, y su metabolito conjugado N-acetilciste\u00edna (SFN-NAC) en las c\u00e9lulas de c\u00e1ncer de pr\u00f3stata.<\/p>\n<p>M\u00c9TODO: SFN y SFN-NAC se analizaron con el modelo de l\u00ednea celular LNCaP de c\u00e1ncer de pr\u00f3stata humano dependiente de andr\u00f3genos. El crecimiento celular y la apoptosis se determinaron con la expresi\u00f3n del receptor de andr\u00f3genos y el ant\u00edgeno prost\u00e1tico espec\u00edfico, la s\u00edntesis de ADN, la progresi\u00f3n del ciclo celular, la rotura de la cadena de ADN y la activaci\u00f3n de caspasa para determinar los efectos y el mecanismo.<\/p>\n<p>RESULTADOS: Se demostr\u00f3 por primera vez que SFN y SFN-NAC median una apoptosis dependiente de la dosis y la detenci\u00f3n del crecimiento en las c\u00e9lulas de c\u00e1ncer de pr\u00f3stata. Se activaron caspasas y se detectaron roturas de cadena de ADN en c\u00e9lulas apopt\u00f3ticas. La expresi\u00f3n de receptores de andr\u00f3genos fosforilados y desfosforilados, y la producci\u00f3n de ant\u00edgeno prost\u00e1tico espec\u00edfico fueron atenuados. La expresi\u00f3n de la ciclina D1 y la s\u00edntesis de ADN fueron inhibidas junto con el bloqueo del ciclo celular G1, causando una disminuci\u00f3n de la densidad celular y el crecimiento. SFN y su metabolito SFN-NAC tienen actividades similares para inducir la detenci\u00f3n del crecimiento y la apoptosis, lo que indica que los efectos de SFN se mantienen a trav\u00e9s de los procesos metab\u00f3licos.<\/p>\n<p>CONCLUSI\u00d3N: Se discute SFN como un componente diet\u00e9tico de vegetales cruc\u00edferos activos en la prevenci\u00f3n del c\u00e1ncer de pr\u00f3stata.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Chiao, J.
