La inhibici\u00f3n de las corrientes de K 2P permite la despolarizaci\u00f3n y excitaci\u00f3n del potencial de membrana. Como se esperaba para los reguladores clave de la excitabilidad, los canales de fuga est\u00e1n bajo un estricto control de una gran cantidad de est\u00edmulos. Recientemente, la se\u00f1alizaci\u00f3n a trav\u00e9s de prote\u00ednas tirosina quinasas (TK) se ha implicado en la modulaci\u00f3n del canal i\u00f3nico. El objetivo de este estudio fue investigar la regulaci\u00f3n de TK de los canales K 2P .<\/p>\n<p>ENFOQUE EXPERIMENTAL: La t\u00e9cnica de fijaci\u00f3n de voltaje de dos electrodos se utiliz\u00f3 para registrar las corrientes de K 2P en los ovocitos de Xenopus . Adem\u00e1s, se estudiaron los canales K 2P en c\u00e9lulas de ovario de h\u00e1mster chino (CHO) utilizando la t\u00e9cnica de pinza de parche de c\u00e9lulas completas.<\/p>\n<p>RESULTADOS CLAVE: Aqu\u00ed, informamos la inhibici\u00f3n de las corrientes humanas K 2P 3.1 (TASK \u2010 1) por el antagonista de TK, geniste\u00edna, en ovocitos de Xenopus (IC 50 = 10.7 \u03bc M ) y en c\u00e9lulas CHO (IC 50 = 12.3 \u03bc M ). El mecanismo molecular subyacente se estudi\u00f3 en detalle. hK 2P 3.1 no fue afectado por la genistina, un an\u00e1logo inactivo de la geniste\u00edna. El perortovanadato, un inhibidor de la actividad de la tirosina fosfatasa, redujo el efecto inhibidor de la geniste\u00edna. La reducci\u00f3n de corriente era independiente del voltaje y no requer\u00eda protonaci\u00f3n de canal en la posici\u00f3n H98 o fosforilaci\u00f3n en el sitio de fosforilaci\u00f3n de TK \u00fanico, Y323.
