El mecanismo celular y molecular de la apoptosis inducida por quercetina se investig\u00f3 en el presente estudio.<\/p>\n<p>M\u00c9TODOS: La morfolog\u00eda nuclear se determin\u00f3 por microscop\u00eda de fluorescencia. La fragmentaci\u00f3n del ADN se analiz\u00f3 por electroforesis y se cuantific\u00f3 por el m\u00e9todo de la difenilamina. El potencial transmembrana de las mitocondrias se midi\u00f3 por citometr\u00eda de flujo. Bcl-2, Bcl-X L , PKC-\u03b1, PKC-\u03b2 y PKC-\u03b4 se detectaron mediante transferencia Western. La actividad de caspasa se determin\u00f3 por espectrofotometr\u00eda.<\/p>\n<p>RESULTADOS: La quercetina indujo la condensaci\u00f3n de los n\u00facleos de las c\u00e9lulas B16-BL6 en un patr\u00f3n dependiente de la dosis, tal como lo visualiza Hoechst 33258 y la muerte por yoduro de propidio. Phorbol 12-miristato 13-acetato (PMA), un activador de PKC, mejor\u00f3 significativamente la apoptosis inducida por quercetina, mientras que la doxorrubicina, un inhibidor de PKC, disminuy\u00f3 notablemente. Tanto la PMA como la doxorrubicina mostraron un efecto constante sobre la fragmentaci\u00f3n del ADN nuclear causado por varias dosis de quercetina.<\/p>\n<p>La quercetina, dependiente de la dosis, condujo a la p\u00e9rdida del potencial de membrana mitocondrial, que tambi\u00e9n se reforz\u00f3 o antagoniz\u00f3 significativamente por PMA y doxorrubicina, respectivamente. Adem\u00e1s, la PMA mostr\u00f3 refuerzo, mientras que la doxorrubicina mostr\u00f3 una antagonizaci\u00f3n significativa de la disminuci\u00f3n mediada por quercetina en la expresi\u00f3n de Bcl-2.<\/p>\n<p>La quercetina promovi\u00f3 la actividad caspasa-3 de una manera dependiente de la dosis, que tambi\u00e9n estaba regulado por PMA y doxorrubicina con un patr\u00f3n similar al observado en su efecto sobre la apoptosis, el potencial de membrana mitocondrial y la expresi\u00f3n de Bcl-2, pero ninguno de estos se vio directamente afectado por PMA y doxorrubicina. El \u00e1cido graso libre y la clorpromazina, un activador e inhibidor de la PKC, respectivamente, no interfirieron con estos efectos de la quercetina. Las c\u00e9lulas B16-BL6 expresaron PKC-\u03b1, PKC-\u03b2 y PKC-\u03b4.<\/p>\n<p>La quercetina inhibi\u00f3 de forma dependiente de la dosis la expresi\u00f3n de PKC-\u03b1 pero no la de PKC-\u03b2 y PKC-\u03b4. La doxorrubicina bloque\u00f3 casi por completo el efecto de la quercetina sobre la expresi\u00f3n de PKC-\u03b1. La quercetina tambi\u00e9n particip\u00f3 en la translocaci\u00f3n de PKC-\u03b4 desde el citosol al n\u00facleo. PMA mejor\u00f3 el efecto de la quercetina en la translocaci\u00f3n de PKC-\u03b4. potencial de membrana mitocondrial y expresi\u00f3n de Bcl-2, pero ninguno de estos se vio directamente afectado por PMA y doxorrubicina. El \u00e1cido graso libre y la clorpromazina, un activador e inhibidor de la PKC, respectivamente, no interfirieron con estos efectos de la quercetina. Las c\u00e9lulas B16-BL6 expresaron PKC-\u03b1, PKC-\u03b2 y PKC-\u03b4.<\/p>\n<p>CONCLUSIONES: Estos resultados indican que la quercetina indujo la apoptosis de las c\u00e9lulas B16-BL6 de melanoma murino al da\u00f1ar sus mitocondrias, aumentar la actividad de la caspasa-3, inhibir la expresi\u00f3n de Bcl-2 y PKC-\u03b1 e inducir la translocaci\u00f3n de PKC-\u03b4. La doxorrubicina inhibi\u00f3 estos efectos de la quercetina al bloquear la disminuci\u00f3n de la expresi\u00f3n de PKC-\u03b1 inducida por la quercetina, mientras que la PMA aument\u00f3 estos efectos al mejorar la translocaci\u00f3n de PKC-\u03b4 inducida por la quercetina.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Zhang, X.
