A una concentraci\u00f3n de 20 \u03bc M , tanto Lu como Qu disminuyeron notablemente los niveles de fosforilaci\u00f3n de las prote\u00ednas celulares A431, incluido el EGFR.<\/p>\n<p>Las c\u00e9lulas A431 tratadas con Lu o Qu exhibieron ampollas citoplasm\u00e1ticas protuberantes y morfolog\u00eda de contracci\u00f3n progresiva. Lu y Qu tambi\u00e9n indujeron de forma dependiente del tiempo la aparici\u00f3n de un patr\u00f3n de escala de fragmentaci\u00f3n de ADN, y este efecto fue abolido por el tratamiento con EGF.<\/p>\n<p>La adici\u00f3n de EGF solo disminuy\u00f3 marginalmente el efecto inhibidor de luteolina y quercetina sobre la tasa de crecimiento de las c\u00e9lulas A431, el tratamiento de prote\u00ednas celulares con EGF y luteolina o quercetina redujo en gran medida la fosforilaci\u00f3n de prote\u00ednas, lo que indica que Lu y Qu pueden actuar eficazmente para inhibir una amplia gama de prote\u00ednas quinasas, incluida la tirosina quinasa EGFR.<\/p>\n<p>EGF aument\u00f3 los niveles de metaloproteinasa matriz 2 (MMP-2) y metaloproteinasa matriz 9 (MMP-9), mientras que Lu y Qu parec\u00edan suprimir la secreci\u00f3n de estas dos MMP en c\u00e9lulas A431.<\/p>\n<p>El examen de la relaci\u00f3n entre la estructura qu\u00edmica y los efectos inhibitorios de ocho flavonoides revela que el doble enlace entre C2 y C3 en el anillo C y los grupos OH en C3 &#8216;y C4&#8217; en el anillo B son cr\u00edticos para las actividades biol\u00f3gicas.<\/p>\n<p>Este estudio demuestra que los efectos inhibitorios de Lu y Qu, y los efectos estimuladores de EGF, sobre la proliferaci\u00f3n de c\u00e9lulas tumorales, la fosforilaci\u00f3n de prote\u00ednas celulares y la secreci\u00f3n de MMP pueden estar mediadas al menos en parte a trav\u00e9s de EGFR.
