Sin embargo, el efecto de la glicosilaci\u00f3n en sus funciones biol\u00f3gicas a\u00fan no est\u00e1 definido.\u00a0<\/p>\n<div>\u00a0<\/div>\n<div>OBJETIVO: En el presente estudio se estudiaron las actividades inductoras de apoptosis de tres flavonoides relacionados con la estructura, entre ellos la quercetina de aglycone (QUE), la rutina de glicona (RUT; QUE-3-O-rutinoside) y la quercitrina de glicona (QUI; QUE-3-O-rhamnoside). Tanto el RUT como el QUI son glic\u00f3sidos QUE, y poseen rutinosa y ramnosa en la posici\u00f3n C3 de QUE, respectivamente.\u00a0<\/div>\n<div>\u00a0<\/div>\n<div>M\u00c9TODOS \/ RESULTADOS: Los resultados del ensayo MTT mostraron que QUE, pero no RUT y QUI, exhibe un efecto citot\u00f3xico significativo en las c\u00e9lulas HL-60, acompa\u00f1ado de la aparici\u00f3n, dependiente de la dosis y el tiempo, de caracter\u00edsticas de apoptosis que incluyen un aumento en la intensidad de la escalera de ADN, cambios morfol\u00f3gicos, cuerpos apopt\u00f3ticos y un aumento de las c\u00e9lulas hipodiploides por an\u00e1lisis de citometr\u00eda de flujo. QUE, pero no RUT o QUI, caus\u00f3 la inducci\u00f3n r\u00e1pida y transitoria de la actividad de la caspasa 3\/CPP32, pero no la actividad de la caspasa 1, de acuerdo con la divisi\u00f3n de los sustratos de la caspasa 3 de la polimerasa (PARP) y las prote\u00ednas D4-GDI, y la aparici\u00f3n de fragmentos de caspasa 3 divididos que se detectaron en las c\u00e9lulas HL-60 tratadas con QUE, pero no con RUT o QUI. Se detect\u00f3 una disminuci\u00f3n de la prote\u00edna anti-apopt\u00f3tica, Mcl-1, en las c\u00e9lulas HL-60 tratadas con QUE, mientras que otras prote\u00ednas de la familia Bcl-2, incluyendo Bax, Bcl-2, Bcl-XL y Bag permanecieron sin cambios. El inhibidor de caspasa 3, Ac-DEVD-FMK, pero no el inhibidor de caspasa 1, Ac-YVAD-FMK, atenu\u00f3 la muerte celular inducida por el QUE. Los resultados del ensayo DCHF-DA indican que no se encontr\u00f3 un aumento significativo en el nivel de per\u00f3xido intracelular en las c\u00e9lulas tratadas con QU\u00c9, y que el QU\u00c9 inhib\u00eda el nivel de per\u00f3xido intracelular inducido por el H(2)O(2). Los depuradores de radicales libres N-acetilciste\u00edna (NAC) y la catalasa no mostraron ninguna prevenci\u00f3n de la apoptosis inducida por el QUE. Adem\u00e1s, la QUE no indujo apoptosis en una l\u00ednea celular monoc\u00edtica madura THP-1, caracterizada por la falta de escalas de ADN, la activaci\u00f3n de la caspasa 3, la divisi\u00f3n PARP y una disminuci\u00f3n de Mcl-1, en comparaci\u00f3n con las c\u00e9lulas HL-60.\u00a0<\/div>\n<div>\u00a0<\/div>\n<div>CONCLUSIONES: Nuestros experimentos proporcionan pruebas que indican que la adici\u00f3n de rutinosa o ramnosa aten\u00faa la actividad inductora de apoptosis de la QUE, y que est\u00e1 implicada la cascada de caspasa 3 pero no la producci\u00f3n de radicales libres.<\/div>\n","protected":false},"excerpt":{"rendered":"<p>Shen, S.
