(2015).<a href=\"https:\/\/www.researchgate.net\/profile\/Claudio_Cabello-Verrugio\/publication\/277011028_Endotoxin-induced_skeletal_muscle_wasting_is_prevented_by_angiotensin_1-7_through_a_p38_MAPK_dependent_mechanism\/links\/568ac86e08ae1e63f1fbf590.pdf\">\u00a0Endotoxin-induced skeletal muscle wasting is prevented by angiotensin-(1\u20137) through a p38 MAPK-dependent mechanism.<\/a>\u00a0<i>Clinical science<\/i>, 129(6), 461-476.<\/p>\n<p>La atrofia del m\u00fasculo esquel\u00e9tico inducida durante el s\u00edndrome de sepsis producido por la endotoxina en forma de LPS (lipopolisac\u00e1rido), es una condici\u00f3n patol\u00f3gica caracterizada por la p\u00e9rdida de fuerza y \u200b\u200bmasa muscular, un aumento en la degradaci\u00f3n del MHC (cadena pesada de miosina) y un aumento en la expresi\u00f3n de atrogin-1 y MuRF-1 (prote\u00edna 1 de dedo RING espec\u00edfica de m\u00fasculo), dos ligasas de ubiquitina E3 que pertenecen al sistema ubiquitina-proteasoma. Ang- (1-7) [Angiotensina- (1-7)], a trav\u00e9s de su receptor Mas, tiene efectos beneficiosos en el m\u00fasculo esquel\u00e9tico.<\/p>\n<p>Evaluamos in vivo el papel del receptor Ang- (1-7) y el receptor Mas en el desgaste muscular inducido por la inyecci\u00f3n de LPS en ratones C57BL \/ 10J. Se realizaron estudios in vitro en miotubos C2C12 murinos y miofibras aisladas del m\u00fasculo EDL (extensor largo de los dedos). Adicionalmente, Se evalu\u00f3 la participaci\u00f3n de p38 MAPK (prote\u00edna quinasa activada por mit\u00f3geno) en el efecto Ang- (1-7) sobre la atrofia muscular inducida por LPS.<\/p>\n<p>Nuestros resultados muestran que Ang- (1-7) previene la disminuci\u00f3n del di\u00e1metro de miofibras y miotubos, la disminuci\u00f3n de la fuerza muscular, la disminuci\u00f3n de los niveles de MHC y la inducci\u00f3n de la expresi\u00f3n de atrogin-1 y MuRF-1, todos los cuales son inducido por LPS. Estos efectos fueron revertidos usando A779, un antagonista de Mas. Ang- (1-7) ejerce estos efectos antiatr\u00f3ficos al menos en parte al inhibir la activaci\u00f3n dependiente de LPS de p38 MAPK tanto in vitro como in vivo.<\/p>\n<p>Hemos demostrado por primera vez que Ang- (1-7) contrarresta la atrofia del m\u00fasculo esquel\u00e9tico inducida por la endotoxina a trav\u00e9s de un mecanismo dependiente del receptor Mas que implica una disminuci\u00f3n en la fosforilaci\u00f3n de p38 MAPK.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Morales, M.
