(2015).\u00a0<a href=\"https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/S1043661815000663\">Ginkgo biloba extract promotes osteogenic differentiation of human bone marrow mesenchymal stem cells in a pathway involving Wnt\/\u03b2-catenin signaling<\/a>.\u00a0<i>Pharmacological Research<\/i>, 97, 70-78.<\/p>\n<p>Las c\u00e9lulas madre mesenquimatosas derivadas de la m\u00e9dula \u00f3sea humana (BM-MSC) son una nueva fuente celular utilizada en la terapia con c\u00e9lulas madre para tratar enfermedades \u00f3seas debido a su alto potencial para diferenciarse en osteoblastos. La inducci\u00f3n efectiva de la diferenciaci\u00f3n osteog\u00e9nica de las BM-MSC humanas es cr\u00edtica para alcanzar su potencial terap\u00e9utico.<\/p>\n<div>\n<p>En este estudio, el extracto de Ginkgo biloba (GBE), una medicina herbal tradicional, se utiliz\u00f3 para estimular la proliferaci\u00f3n y la diferenciaci\u00f3n osteog\u00e9nica de las BM-MSC humanas. El presente estudio revel\u00f3 que GBE mejor\u00f3 la proliferaci\u00f3n y la osteog\u00e9nesis de las BM-MSC humanas de una manera dependiente de la dosis en el rango de 25-75 mg \/ l, como lo indica la actividad de fosfatasa alcalina (ALP) y el contenido de calcio. Sin embargo, dicho efecto disminuy\u00f3 o inhibi\u00f3 a 100 mg \/ lo m\u00e1s alto.<\/p>\n<p>La mejora dependiente de la dosis en la osteog\u00e9nesis de las BM-MSC humanas por GBE se confirm\u00f3 a\u00fan m\u00e1s por la regulaci\u00f3n al alza dependiente de la dosis de genes marcadores, osteopontina (OPN) y col\u00e1geno I. La expresi\u00f3n incrementada de osteoprotegerina (OPG) y la expresi\u00f3n m\u00ednima del activador del receptor nuclear el ligando de factor-B (RANKL) sugiri\u00f3 que GBE tambi\u00e9n inhibi\u00f3 la osteoclastog\u00e9nesis de BM-MSC humanas. Un estudio mecanicista posterior demostr\u00f3 que los niveles transcripcionales de la prote\u00edna morfogen\u00e9tica \u00f3sea 4 (BMP4) y el factor de transcripci\u00f3n 2 relacionado con el Runt (RUNX2) en la se\u00f1alizaci\u00f3n de BMP, \u03b2-catenina y Ciclina D1en la se\u00f1alizaci\u00f3n de Wnt \/ \u03b2-catenina, aument\u00f3 significativamente durante la osteog\u00e9nesis promovida por GBE.<\/p>\n<p>Mientras tanto, el ensayo de p\u00e9rdida de funci\u00f3n con los inhibidores de se\u00f1alizaci\u00f3n confirm\u00f3 que las v\u00edas de se\u00f1alizaci\u00f3n BMP y Wnt \/ \u03b2-catenina eran indispensables durante la osteog\u00e9nesis promovida por GBE, lo que sugiere que GBE mejor\u00f3 la osteog\u00e9nesis a trav\u00e9s de la regulaci\u00f3n positiva de BMP y Wnt \/ \u03b2 -catenina de se\u00f1alizaci\u00f3n.<\/p>\n<p>El presente estudio propuso que GBE se usara para aumentar la diferenciaci\u00f3n osteog\u00e9nica de BM-MSC humanas para la formaci\u00f3n de hueso nuevo en la terapia celular basada en BM-MSC, lo que podr\u00eda proporcionar un tratamiento atractivo y prometedor para los trastornos \u00f3seos.<\/p>\n<\/div>\n","protected":false},"excerpt":{"rendered":"<p>Gu, Q., Chen, C., Zhang, Z., Wu, Z., Fan, X., Zhang, Z., &#8230; &amp; Shi, L.
