Como la interleucina-1\u03b2 (IL-1\u03b2) es una de las citocinas proinflamatorias clave que contribuyen a la progresi\u00f3n en la OA, investigamos el efecto de Se en la neutralizaci\u00f3n de los efectos inflamatorios de la IL-1\u03b2 en el \u00f3xido n\u00edtrico (NO) y la prostaglandina E\u2082 (PGE\u2082) producci\u00f3n y las v\u00edas de se\u00f1alizaci\u00f3n involucradas.<\/p>\n<p>M\u00c9TODOS: Los condrocitos humanos primarios aislados se pretrataron con selenometionina (SeMet) (SeMet 0,5 \u03bcM) durante 24 h, luego se trataron conjuntamente sin o con IL-1\u03b2 (10 pg \/ ml o 50 pg \/ ml) durante otras 24 h seguido de aislamiento de ARN . La expresi\u00f3n g\u00e9nica de \u00f3xido n\u00edtrico sintasa inducible (iNOS) y ciclooxigenasa-2 (COX2) se determin\u00f3 mediante reacci\u00f3n en cadena cuantitativa en tiempo real de la polimerasa. Las concentraciones en los medios de cultivo de NO y PGE\u2082 se determinaron por ensayo de nitrito (NO\u2082\u207b) e inmunoensayo, respectivamente. Para el an\u00e1lisis de las v\u00edas de se\u00f1alizaci\u00f3n celular, los condrocitos se pretrataron con SeMet y luego se estimularon con IL-1\u03b2 durante 0-45 min. La actividad de las rutas de se\u00f1alizaci\u00f3n de IL-1\u03b2 se determin\u00f3 mediante detecci\u00f3n por transferencia Western de estados de fosforilaci\u00f3n de prote\u00ednas de transducci\u00f3n de se\u00f1ales.<\/p>\n<p>RESULTADOS: SeMet inhibi\u00f3 la expresi\u00f3n del gen de condrocitos de iNOS inducida por IL-1\u03b2 (31-54%, P = 0.031) y COX2 (50-65%, P = 0.031) con las reducciones correspondientes en ambos NO (19-47%, P = 0.031 ) y PGE\u2082 (24-32%, P = 0.031) producci\u00f3n. El pretratamiento con SeMet atenu\u00f3 la activaci\u00f3n inducida por IL-1\u03b2 de p38 MAPK (39%, P = 0.039) pero no las v\u00edas de quinasa reguladas por se\u00f1al extracelular (ERK) 1\/2, c-Jun quinasas N-terminales (JNK) o factor nuclear \u03baB (NF\u03baB).<\/p>\n<p>CONCLUSIONES: Este estudio aclara un posible mecanismo protector de Se, a trav\u00e9s de la alteraci\u00f3n de la se\u00f1alizaci\u00f3n celular y la transcripci\u00f3n posterior de los efectos proinflamatorios de IL-1\u03b2.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Cheng, A.
