Journal of Virology, 91(21).<\/p>\n<p>CONTEXTO: El virus de la hepatitis E (HEV) causa una hepatitis aguda y autolimitada en individuos sanos y conduce a enfermedades cr\u00f3nicas en individuos inmunocomprometidos. La infecci\u00f3n por HEV en mujeres embarazadas produce un resultado m\u00e1s grave, con una tasa de mortalidad de hasta el 30%. Aunque el virus generalmente causa infecci\u00f3n espor\u00e1dica, se han reportado epidemias en pa\u00edses en desarrollo y privados de recursos. No existe un antiviral espec\u00edfico contra el HEV. Se ha utilizado una combinaci\u00f3n de terapia con interfer\u00f3n y ribavirina para controlar la enfermedad con cierto \u00e9xito. El zinc es un micronutriente esencial que juega un papel crucial en m\u00faltiples procesos celulares. Se sabe que las sales de zinc son efectivas para reducir las infecciones causadas por pocos virus.<\/p>\n<p>OBJETIVO: Aqu\u00ed, investigamos el efecto de las sales de zinc en la replicaci\u00f3n de HEV.<\/p>\n<p>M\u00c9TODO \/ RESULTADOS: En un modelo de cultivo de c\u00e9lulas de hepatoma humano (Huh7), las sales de zinc inhibieron la replicaci\u00f3n de los replicones de genotipo 1 (g-1) y g-3 HEV y el ARN gen\u00f3mico infeccioso g-1 HEV de una manera dependiente de la dosis. El an\u00e1lisis de un mutante defectuoso en la replicaci\u00f3n del ARN gen\u00f3mico de HEV g-1 en condiciones similares descart\u00f3 la posibilidad de que las sales de zinc act\u00faen en procesos independientes de la replicaci\u00f3n. Un modelo de infecci\u00f3n basada en la l\u00ednea celular ORF4-Huh7 de HEV g-1 confirm\u00f3 a\u00fan m\u00e1s las observaciones anteriores. Las sales de zinc no mostraron ning\u00fan efecto sobre la entrada de g-1 HEV en la c\u00e9lula hu\u00e9sped. Adem\u00e1s, nuestros datos revelan que las sales de zinc inhiben directamente la actividad de la ARN polimerasa dependiente de ARN viral (RdRp), lo que conduce a la inhibici\u00f3n de la replicaci\u00f3n viral.<\/p>\n<p>CONCLUSI\u00d3N: Tomados en conjunto, estos estudios desentra\u00f1an la capacidad de las sales de zinc para inhibir la replicaci\u00f3n de HEV, lo que sugiere su posible valor terap\u00e9utico para controlar la infecci\u00f3n por HEV.<\/p>\n<p>IMPORTANCIA: El virus de la hepatitis E (HEV) es un problema de salud p\u00fablica en los pa\u00edses que carecen de recursos debido a los brotes frecuentes. Tambi\u00e9n est\u00e1 surgiendo como un problema de salud en los pa\u00edses desarrollados debido a su capacidad de causar infecci\u00f3n aguda y cr\u00f3nica en trasplantes de \u00f3rganos e individuos inmunocomprometidos. Aunque los antivirales como la ribavirina se han utilizado para tratar los casos de HEV, se conocen efectos secundarios y limitaciones de dicha terapia. Nuestro descubrimiento de la capacidad de las sales de zinc para bloquear la replicaci\u00f3n de HEV en virtud de su capacidad de inhibir la actividad de RdRp viral es importante porque estos hallazgos allanan el camino para probar la eficacia de la terapia de suplementaci\u00f3n con zinc en pacientes infectados con HEV.
