La angiog\u00e9nesis requiere una degradaci\u00f3n de la matriz extracelular estrechamente controlada mediada por enzimas proteol\u00edticas extracelulares, incluidas las metaloproteinasas de la matriz (MMP) y las serina proteasas, en particular, el sistema activador de plasmin\u00f3geno de tipo uroquinasa (uPA).<\/p>\n<p>OBJETIVO \/ M\u00c9TODO: En este estudio, hemos investigado el mecanismo antiangiog\u00e9nico de los flavonoides, geniste\u00edna, apigenina y 3-hidroxiflavona en un modelo de c\u00e9lulas endoteliales de la vena umbilical humana (HUVEC). La estimulaci\u00f3n de HUVEC sin suero con factor de crecimiento endotelial vascular \/ factor de crecimiento b\u00e1sico de fibroblastos (VEGF \/ bFGF) caus\u00f3 un marcado aumento en la producci\u00f3n de MMP-1 e indujo la activaci\u00f3n pro-MMP-2 acompa\u00f1ada por un aumento en la expresi\u00f3n de MT1-MMP.<\/p>\n<p>RESULTADOS: Sin embargo, el pretratamiento con flavonoides antes de la estimulaci\u00f3n de VEGF \/ bFGF aboli\u00f3 por completo el aumento estimulado por VEGF \/ bFGF en la expresi\u00f3n de MMP \u2010 1 y MT1 \u2010 MMP y la activaci\u00f3n pro \u2010 MMP \u2010 2. La geniste\u00edna bloque\u00f3 el aumento estimulado por VEGF \/ bFGF en la expresi\u00f3n de TIMP-1 y la disminuci\u00f3n en la expresi\u00f3n de TIMP-2. La apigenina y la 3-hidroxiflavona disminuyeron a\u00fan m\u00e1s la expresi\u00f3n de TIMP-1 por debajo del nivel basal y abolieron por completo la expresi\u00f3n de TIMP-2. La estimulaci\u00f3n de VEGF y bFGF tambi\u00e9n indujo significativamente la expresi\u00f3n de uPA, m\u00e1s notablemente el nivel de uPA de 33 kDa, y aument\u00f3 la expresi\u00f3n del inhibidor de PA (PAI) \u20101. Geniste\u00edna, apigenina, y 3-hidroxiflavona bloque\u00f3 efectivamente la generaci\u00f3n de uPA de 33 kDa, y disminuy\u00f3 a\u00fan m\u00e1s la actividad de la uPA de 55 kDa y la expresi\u00f3n de PAI-1 por debajo del nivel basal.<\/p>\n<p>CONCLUSI\u00d3N: En conclusi\u00f3n, estos datos sugieren que la geniste\u00edna, la apigenina y la 3-hidroxiflavona inhiben la angiog\u00e9nesis in vitro, en parte mediante la prevenci\u00f3n de la expresi\u00f3n de MMP-1 y uPA inducida por VEGF \/ bFGF y la activaci\u00f3n de pro-MMP-2, y mediante la modulaci\u00f3n de sus inhibidores , TIMP \u2010 1 y \u20102, y PAI \u2010 1.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Kim, M.
