(2017).\u00a0<a href=\"https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/S0378111917301348\">The herbal medicine Melissa officinalis extract effects on gene expression of p53, Bcl-2, Her2, VEGF-A and hTERT in human lung, breast and prostate cancer cell lines.<\/a>\u00a0<i>Gene<\/i>, 613, 14-19.<\/p>\n<p>INTRODUCCI\u00d3N: Anteriormente, verificamos que el extracto de Melissa officinalis (MOE) provoca potentes efectos antiproliferativos en diferentes c\u00e9lulas cancerosas humanas. Para obtener informaci\u00f3n sobre los mecanismos moleculares que explican los efectos citot\u00f3xicos de MOE, evaluamos los patrones de expresi\u00f3n de varias mol\u00e9culas prominentes con potencial terap\u00e9utico en el c\u00e1ncer mediante PCR cuantitativa (Q-PCR).<\/p>\n<p>M\u00c9TODOS: Las c\u00e9lulas cancerosas A549, MCF-7 y PC3 se cultivaron en RPMI 1640 completo y se sembraron en microplacas de 24 pocillos. Despu\u00e9s de la incubaci\u00f3n durante 72 h, se a\u00f1adieron 100 \u03bcg \/ ml de MOE y las c\u00e9lulas se incubaron adicionalmente durante 72 h. Posteriormente, las c\u00e9lulas se sometieron a extracci\u00f3n de ARN para los medios de Q-PCR.<\/p>\n<p>RESULTADOS: Nuestros resultados indicaron que en las c\u00e9lulas cancerosas PC3, MOE result\u00f3 en una regulaci\u00f3n negativa significativa de VEGF-A (0.0004 veces), Bcl-2 (0.001 veces), Her2 (0.02 veces) y hTERT (0.023 veces) en comparaci\u00f3n con el control no tratado. Adem\u00e1s, el ARNm de VEGF-A y hTERT tambi\u00e9n se redujo significativamente en las c\u00e9lulas cancerosas MCF-7 y A549. En particular, la alta actividad antiangiog\u00e9nica se asoci\u00f3 estrechamente con una alta actividad anti-telomerasa de MOE en el estudio de las c\u00e9lulas cancerosas. La disminuci\u00f3n en la expresi\u00f3n de VEGF-A fue significativamente superior a la de la regulaci\u00f3n por disminuci\u00f3n de hTERT, ya que las c\u00e9lulas cancerosas PC3 con la mayor regulaci\u00f3n por disminuci\u00f3n de hTERT (0.023) presentaron la mayor anti VEGFactividad (0.0004 veces), mientras que las c\u00e9lulas MCF-7 con la inhibici\u00f3n de hTERT m\u00e1s baja (0.213) mostraron la inhibici\u00f3n de VEGF m\u00e1s baja (0.0435) entre las tres c\u00e9lulas cancerosas estudiadas. Notamos que la modulaci\u00f3n de la expresi\u00f3n g\u00e9nica de VEGF-A y hTERT puede considerarse como un objetivo com\u00fan, lo que explica el potencial terap\u00e9utico de MOE en c\u00e9lulas de c\u00e1ncer de mama, pulm\u00f3n y pr\u00f3stata humanos.<\/p>\n<p>CONCLUSI\u00d3N: En conjunto, se sugiere que la potente actividad antiproliferativa del extracto hidroalcoh\u00f3lico de Melissa officinalis se explica de alguna manera por su alta potencia para inhibir la expresi\u00f3n de los oncogenes prominentes Bcl2, Her2, VEGF-A y hTERT en el c\u00e1ncer de pr\u00f3stata. En los tumores con p53 funcional, incluidas las c\u00e9lulas cancerosas MCF-7 y A549, el papel de p53, Bcl2 y Her2 es menos significativo. Parece que el MOE ejerce sus efectos antiproliferativos en estas c\u00e9lulas cancerosas, en parte a trav\u00e9s de la regulaci\u00f3n a la baja concurrente de VEGF-A y hTERT.
