C<i>ancer chemotherapy and pharmacology<\/i>, 79(5), 1031-1041.<\/p>\n<p>OBJETIVO: La luteolina, un flavonoide diet\u00e9tico com\u00fan, induce la apoptosis de muchos tipos de c\u00e9lulas cancerosas. Sin embargo, se desconoce su papel en el glioblastoma y los posibles mecanismos. En esta investigaci\u00f3n, estudiamos los mecanismos moleculares del efecto anticanceroso de la luteolina en l\u00edneas celulares de c\u00e1ncer de glioblastoma.<\/p>\n<p>M\u00c9TODOS: Se ensayaron las l\u00edneas celulares de glioblastoma humano U251MG y U87MG. El crecimiento celular se evalu\u00f3 mediante el kit de recuento celular-8. La apoptosis celular se detect\u00f3 con citometr\u00eda de flujo y tinci\u00f3n con inmunofluorescencia de caspasa-3. Los niveles de prote\u00edna de la ruta caspasa-3 \/ Bax \/ Bcl-2 y p-PERK \/ p-eIF2\u03b1 \/ ATF4 \/ CHOP \/ caspasa-12 se analizaron utilizando transferencias de Western. La generaci\u00f3n de especies reactivas de ox\u00edgeno se midi\u00f3 con tinci\u00f3n DCFH-DA usando citometr\u00eda de flujo. El potencial de membrana mitocondrial se prob\u00f3 con tinci\u00f3n JC-1. El efecto anticanceroso in vivo se midi\u00f3 usando el modo de xenoinjerto tumoral en ratones desnudos.<\/p>\n<p>RESULTADOS: La luteolina indujo una respuesta letal al estr\u00e9s del ret\u00edculo endopl\u00e1smico y disfunci\u00f3n mitocondrial en las c\u00e9lulas del glioblastoma al aumentar los niveles de especies reactivas de ox\u00edgeno (ROS) intracelulares. La luteolina indujo la expresi\u00f3n de prote\u00ednas asociadas al estr\u00e9s del RE, incluida la fosforilaci\u00f3n de PERK, eIF2\u03b1, ATF4, CHOP y caspasa escindida 12. La inhibici\u00f3n de la producci\u00f3n de ROS por el antioxidante N- acetilciste\u00edna podr\u00eda revertir el estr\u00e9s del RE inducido por luteolina y la activaci\u00f3n de las v\u00edas mitocondriales tambi\u00e9n como apoptosis. Adem\u00e1s, tambi\u00e9n mostramos el efecto anticanceroso de la luteolina in vivo.<\/p>\n<p>CONCLUSIONES: Nuestros resultados sugieren que la luteolina induce la apoptosis activando el estr\u00e9s del RE y la disfunci\u00f3n mitocondrial en l\u00edneas celulares de glioblastoma e in vivo, lo que proporciona el candidato anticancer\u00edgeno para tratar el glioblstoma.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Wang, Q., Wang, H., Jia, Y., Pan, H., &amp; Ding, H.
