(2002).\u00a0<a href=\"https:\/\/www.jbc.org\/content\/277\/3\/1705.full.pdf\">Polyunsaturated fatty acids suppress sterol regulatory element-binding protein 1c promoter activity by inhibition of liver X receptor (LXR) binding to LXR response elements<\/a>.\u00a0<i>Journal of Biological Chemistry<\/i>, 277(3), 1705-1711.<\/p>\n<p>FONDO: Estudios anteriores han demostrado que los \u00e1cidos grasos poliinsaturados (PUFA) suprimen la expresi\u00f3n de la prote\u00edna de uni\u00f3n a elementos reguladores de esterol 1c (SREBP-1c) y, por tanto, la lipog\u00e9nesis.<\/p>\n<p>OBJETIVO \/ METODOLOG\u00cdA: En el estudio actual, el mecanismo molecular de este efecto supresor se investig\u00f3 con ensayos del gen indicador de luciferasa utilizando el promotor SREBP-1c en c\u00e9lulas HEK293.<\/p>\n<p>RESULTADOS: De acuerdo con los datos anteriores, la adici\u00f3n de PUFA al medio en los ensayos inhibi\u00f3 de manera contundente la actividad del promotor SREBP-1c. La deleci\u00f3n y mutaci\u00f3n de los dos elementos que responden al receptor X del h\u00edgado (LXR) (LXRE) en la regi\u00f3n promotora de SREBP-1c elimin\u00f3 este efecto supresor, lo que indica que ambos LXRE son elementos supresores de PUFA importantes. Las actividades de luciferasa tanto del promotor SREBP-1c como de las construcciones potenciadoras de LXRE inducidas por la coexpresi\u00f3n de LXRalpha o -beta fueron fuertemente suprimidas por la adici\u00f3n de varios PUFA (\u00e1cido araquid\u00f3nico&gt; \u00e1cido eicosapentaenoico&gt; \u00e1cido docosahexaenoico&gt; \u00e1cido linoleico), mientras que el \u00e1cido saturado o mono -los \u00e1cidos grasos insaturados tuvieron efectos m\u00ednimos.
