(2010).\u00a0<a href=\"http:\/\/journals.plos.org\/plosone\/article?id=10.1371\/journal.pone.0011457\">Sulforaphane causes epigenetic repression of hTERT expression in human breast cancer cell lines<\/a>.\u00a0<i>PloS one<\/i>, 5(7), e11457.<\/p>\n<p>ANTECEDENTES: El sulforafano (SFN), un isotiocianato que se encuentra en las verduras cruc\u00edferas, es un componente diet\u00e9tico com\u00fan que tiene actividad inhibidora de la histona desacetilasa y un potencial emocionante en la prevenci\u00f3n del c\u00e1ncer. Los mecanismos por los cuales SFN imparte sus propiedades quimiopreventivas son de considerable inter\u00e9s y se sabe poco sobre su potencial preventivo para el c\u00e1ncer de mama.<\/p>\n<p>PRINCIPALES HALLAZGOS: Descubrimos que SFN inhibe significativamente la viabilidad y la proliferaci\u00f3n de las c\u00e9lulas de c\u00e1ncer de mama in vitro, mientras que tiene efectos insignificantes en las c\u00e9lulas de mama normales. La inhibici\u00f3n de la telomerasa ha recibido una atenci\u00f3n considerable debido a su alta expresi\u00f3n en c\u00e9lulas cancerosas y su nivel extremadamente bajo de expresi\u00f3n en c\u00e9lulas normales. El tratamiento con SFN inhibi\u00f3 de forma dependiente de la dosis y del tiempo la transcriptasa inversa de la telomerasa humana ( hTERT ), la subunidad reguladora catal\u00edtica de la telomerasa, en las c\u00e9lulas de c\u00e1ncer de mama humano MCF-7 y MDA-MB-231. Las metiltransferasas de ADN (DNMT), especialmente DNMT1 y DNMT3a, tambi\u00e9n disminuyeron en las c\u00e9lulas de c\u00e1ncer de mama tratadas con SFN, lo que sugiere que SFN puede reprimir la hTERT.impactando las v\u00edas epigen\u00e9ticas. Regulaci\u00f3n descendente de DNMT en respuesta a la desmetilaci\u00f3n de CpG espec\u00edfica de sitio inducida por SFN que se produce principalmente en el primer ex\u00f3n del gen hTERT , lo que facilita la uni\u00f3n de CTCF asociada con la represi\u00f3n de hTERT . El an\u00e1lisis de inmunoprecipitaci\u00f3n de cromatina (ChIP) del promotor hTERT revel\u00f3 que SFN aument\u00f3 el nivel de marcadores de cromatina activos acetil-H3, acetil-H3K9 y acetil-H4, mientras que los marcadores de cromatina inactivos trimetil-H3K9 y trimetil-H3K27 disminuyeron en una dosis- manera dependiente. La hiperacetilaci\u00f3n inducida por SFN facilit\u00f3 la uni\u00f3n de muchas prote\u00ednas represoras de hTERT como MAD1 y CTCF a la hTERTregi\u00f3n reguladora. El agotamiento de CTCF usando ARNip redujo la regulaci\u00f3n a la baja inducida por SFN de la transcripci\u00f3n del ARNm de hTERT en estas c\u00e9lulas de c\u00e1ncer de mama. Adem\u00e1s, la regulaci\u00f3n a la baja de la expresi\u00f3n de hTERT facilit\u00f3 la inducci\u00f3n de apoptosis celular en c\u00e9lulas de c\u00e1ncer de mama humano.<\/p>\n<p>SIGNIFICADO: En conjunto, nuestros resultados proporcionan conocimientos novedosos sobre la regulaci\u00f3n negativa epigen\u00e9tica de la telomerasa mediada por SFN en la prevenci\u00f3n del c\u00e1ncer de mama y pueden abrir nuevas v\u00edas para los enfoques de prevenci\u00f3n del c\u00e1ncer mediada por SFN.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Meeran, S.
