(2008).\u00a0<a href=\"file:\/\/\/C:\/Users\/Pc\/Documents\/Manuel\/Documents\/MIN\/v\">tabAnti-HER2 (erb B-2) oncogene effects of phenolic compounds directly isolated from commercial Extra-Virgin Olive Oil (EVOO).\u00a0<\/a><i>Bmc Cancer<\/i>, 8(1), 1-23.<\/p>\n<p>ANTECEDENTES: Los efectos de la dieta mediterr\u00e1nea rica en aceite de oliva sobre el riesgo de c\u00e1ncer de mama pueden subestimarse cuando se consideran juntos los carcinomas de mama oncog\u00e9nicos positivos para HER2 ( ERB B2) y negativos para HER2. Aqu\u00ed investigamos los efectos anti-HER2 de las fracciones fen\u00f3licas extra\u00eddas directamente del aceite de oliva virgen extra (AOVE) en l\u00edneas celulares de c\u00e1ncer de mama humano cultivadas.<\/p>\n<p>M\u00c9TODOS: Se utiliz\u00f3 extracci\u00f3n en fase s\u00f3lida seguida de cromatograf\u00eda l\u00edquida de alta resoluci\u00f3n (HPLC) semipreparativa para aislar las fracciones fen\u00f3licas del AOVE comercial. Se realiz\u00f3 electroforesis anal\u00edtica capilar acoplada a espectrometr\u00eda de masas para comprobar la composici\u00f3n y confirmar la identidad de las fracciones aisladas. Las fracciones polifen\u00f3licas de AOVE se probaron en cuanto a su capacidad tumoricida contra modelos in vitro de c\u00e1ncer de mama HER2 negativos y HER2 positivos utilizando MTT, tinci\u00f3n con violeta cristal y ensayos ELISA de muerte celular. Los efectos de las fracciones polifen\u00f3licas de AOVE sobre la expresi\u00f3n y el estado de activaci\u00f3n de la oncoprote\u00edna HER2 se evaluaron mediante ELISA espec\u00edficos de HER2 y procedimientos de inmunotransferencia, respectivamente.<\/p>\n<p>RESULTADOS: Entre las fracciones que contienen principalmente los fenoles individuales hidroxitirosol y tirosol, el \u00e1cido polifen\u00f3lico \u00e1cido elen\u00f3lico, los lignanos (+) &#8211; pinoresinol y 1 &#8211; (+) &#8211; acetoxipinoresinol, y los secoiridoides desacetoxi oleurope\u00edna aglicona, ligstrosida aglicona y oleurope\u00edna aglicona, todos los principales polifenoles de AOVE ( es decirsecoiridoides y lignanos) inducen fuertes efectos tumoricidas dentro de un rango micromolar al desencadenar selectivamente altos niveles de muerte celular apopt\u00f3tica en sobreexpresores de HER2. El peque\u00f1o agotamiento inducido por ARN de interferencia de la prote\u00edna HER2 y el bloqueo inducido por lapatinib de la actividad tirosina quinasa de HER2 evitaron significativamente la citotoxicidad inducida por polifenoles del AOVE. Los polifenoles de AOVE agotaron dr\u00e1sticamente la prote\u00edna HER2 y redujeron la autofosforilaci\u00f3n de tirosina HER2 de una manera dependiente de la dosis y el tiempo. La regulaci\u00f3n a la baja de HER2 inducida por polifenoles de AOVE se produjo independientemente del mecanismo molecular que contribuye a la sobreexpresi\u00f3n de HER2 ( es decir, de forma natural por amplificaci\u00f3n g\u00e9nica e impulsada ect\u00f3picamente por un promotor viral). El pretratamiento con el inhibidor del proteasoma MG132 previno el agotamiento de HER2 inducido por polifenoles del AOVE.<\/p>\n<p>CONCLUSI\u00d3N: La capacidad de los polifenoles derivados del AOVE para inhibir la actividad de HER2 al promover la degradaci\u00f3n proteasomal de la prote\u00edna HER2 en s\u00ed, junto con el hecho de que los humanos han estado ingiriendo secoiridoides y lignanos de manera segura desde que consumieron aceitunas y OO, respaldan la idea de que la estereoqu\u00edmica de estos fitoqu\u00edmicos podr\u00eda proporcionar una plataforma excelente y segura para el dise\u00f1o de nuevos agentes dirigidos a HER2.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Menendez, J.
