Presenta efectos antiproliferativos en las c\u00e9lulas de c\u00e1ncer de mama humano.<\/p>\n<p>Aqu\u00ed examinamos varias l\u00edneas celulares de c\u00e1ncer de mama humano con diferentes niveles de expresi\u00f3n de HER2\/neu y encontramos que la apigenina exhib\u00eda una potente actividad inhibidora del crecimiento en las c\u00e9lulas de c\u00e1ncer de mama que expresaban el HER2\/neu, pero era mucho menos eficaz para las c\u00e9lulas que expresaban niveles basales de HER2\/neu.<\/p>\n<p>Tambi\u00e9n se observ\u00f3 una inducci\u00f3n de apoptosis en las c\u00e9lulas cancerosas de mama que expresaban el HER2\/neu en forma dependiente de la dosis y el tiempo. Sin embargo, quedaban por dilucidar uno o m\u00e1s mecanismos moleculares de la apoptosis inducida por la apigenina en las c\u00e9lulas cancerosas de mama que expresan el HER2\/neu. Se sabe que una v\u00eda de supervivencia celular que involucra a la fosfatidilinositol 3-quinasa (PI3K) y a la Akt juega un papel importante en la inhibici\u00f3n de la apoptosis en respuesta a las c\u00e9lulas cancerosas de mama HER2\/neu-expresadas, lo que nos llev\u00f3 a investigar si esta v\u00eda juega un papel en la apoptosis inducida por la apigenina en las c\u00e9lulas cancerosas de mama HER2\/neu-expresadas.<\/p>\n<p>Nuestros resultados mostraron que la apigenina inhibe la funci\u00f3n de la Akt en las c\u00e9lulas tumorales de una manera compleja. Primero, la apigenina inhibi\u00f3 directamente la actividad del PI3K mientras que indirectamente inhibi\u00f3 la actividad de la quinasa Akt. Segundo, tambi\u00e9n se observ\u00f3 la inhibici\u00f3n de la autofosforilaci\u00f3n y la transfosforilaci\u00f3n del HER2\/neu como resultado del agotamiento de la prote\u00edna HER2\/neu in vivo. Adem\u00e1s, la apigenina inhibi\u00f3 la actividad de la Akt quinasa al impedir el acoplamiento del PI3K a los heterod\u00edmeros HER2\/HER3.<\/p>\n<p>Por lo tanto, propusimos que los efectos celulares inducidos por la apigenina son el resultado de la p\u00e9rdida de la expresi\u00f3n de HER2\/neu y HER3 con la subsiguiente inactivaci\u00f3n de PI3K y AKT en las c\u00e9lulas que dependen de esta v\u00eda para la proliferaci\u00f3n celular y la inhibici\u00f3n de la apoptosis.<\/p>\n<p>Esto implica que la inhibici\u00f3n de la funci\u00f3n del heterod\u00edmero HER2\/HER3 proporcion\u00f3 una estrategia especialmente eficaz para bloquear la transformaci\u00f3n mediada por el HER2\/neu de las c\u00e9lulas de c\u00e1ncer de mama.<\/p>\n<p>Nuestros resultados tambi\u00e9n demostraron que la apigenina disociaba el complejo de HER2\/neu y el GRP94 que preced\u00eda al agotamiento del HER2\/neu.
