En este estudio, cuantificamos e interpretamos los cambios en los patrones de expresi\u00f3n gen\u00e9tica global en la gl\u00e1ndula prost\u00e1tica humana antes, durante y despu\u00e9s de una dieta rica en br\u00f3coli de 12 meses.<\/p>\n<p>METODOS Y HALLAZGOS: Los voluntarios fueron asignados al azar a una dieta rica en br\u00f3coli o en guisantes. Despu\u00e9s de seis meses, no hubo diferencias en la expresi\u00f3n g\u00e9nica entre los individuos con glutati\u00f3n S-transferasa mu 1 ( GSTM1 ) positivos y nulos en la dieta rica en guisantes, pero diferencias significativas entre los genotipos GSTM1 en la dieta rica en br\u00f3coli, asociadas con el factor de crecimiento transformante beta 1 (TGF\u03b21) y v\u00edas de se\u00f1alizaci\u00f3n del factor de crecimiento epid\u00e9rmico (EGF). Comparaci\u00f3n de biopsias obtenidas pre y postLa intervenci\u00f3n revel\u00f3 que se produjeron m\u00e1s cambios en la expresi\u00f3n gen\u00e9tica en individuos con una dieta rica en br\u00f3coli que en aquellos con una dieta rica en guisantes. Si bien hubo cambios en la se\u00f1alizaci\u00f3n de andr\u00f3genos, independientemente de la dieta, los hombres que siguieron la dieta de br\u00f3coli tuvieron cambios adicionales en el procesamiento del ARNm y en la se\u00f1alizaci\u00f3n de TGF\u03b21, EGF e insulina. Tambi\u00e9n proporcionamos evidencia de que el sulforafano (el isotiocianato derivado del glucosinolato de 4-metilsufinilbutilo que se acumula en el br\u00f3coli) interact\u00faa qu\u00edmicamente con TGF\u03b21, EGF y p\u00e9ptidos de insulina para formar tioureas y mejora la transcripci\u00f3n mediada por TGF\u03b21 \/ Smad.<\/p>\n<p>CONCLUSIONES: Estos hallazgos sugieren que el consumo de br\u00f3coli interact\u00faa con el genotipo GSTM1 para producir cambios complejos en las v\u00edas de se\u00f1alizaci\u00f3n asociadas con la inflamaci\u00f3n y la carcinog\u00e9nesis en la pr\u00f3stata. Proponemos que estos cambios pueden estar mediados por la interacci\u00f3n qu\u00edmica de isotiocianatos con p\u00e9ptidos de se\u00f1alizaci\u00f3n en el plasma.
