Wasser et al.(Agaricomycetideae) activates macrophages through toll-like receptor 4<\/a>.\u00a0<i>International Journal of Medicinal Mushrooms<\/i>, 8(3).<\/p>\n<p>Se estimularon macr\u00f3fagos peritoneales de ratones con receptor toll-like (TLR) 4 y TLR2 knockout (TLR4 &#8211; \/ &#8211; y TLR2 &#8211; \/ -) con un heteroglicano insoluble en agua (ABHG), purificado de un cuerpo frutal de Agaricus brasiliensis.<\/p>\n<p>Los ratones TLR4 &#8211; \/ &#8211; no produjeron factor de necrosis tumoral (TNF) -\u03b1 ni \u00f3xido n\u00edtrico (NO), pero TLR2 &#8211; \/ &#8211; s\u00ed. Sin embargo, ABHG no inhibi\u00f3 la producci\u00f3n de TNF-\u03b1 y NO por los macr\u00f3fagos en presencia de polimixina B, que podr\u00eda unirse al l\u00edpido A en el lipopolisac\u00e1rido (LPS); sugiriendo que ABHG no conten\u00eda LPS como contaminantes.<\/p>\n<p>Estos resultados indicaron que ABHG fue reconocido a trav\u00e9s de TLR4 &#8211; \/ &#8211; para producir citocinas inflamatorias como TNF-\u03b1 y NO. Se utiliz\u00f3 RT-PCR para investigar el efecto de ABHG en la expresi\u00f3n de ARNm de TNF-\u03b1 y \u00f3xido n\u00edtrico sintasa inducible (iNOS) en RAW 264.7. La expresi\u00f3n de ARNm de TNF-\u03b1 se indujo 0,5 horas despu\u00e9s de la estimulaci\u00f3n, alcanz\u00f3 su punto m\u00e1ximo en 3 horas y luego disminuy\u00f3 para alcanzar un nivel estable despu\u00e9s de 24 horas.<\/p>\n<p>Por otro lado, la inducci\u00f3n de ARNm de iNOS se retras\u00f3 durante 3 horas y, posteriormente, se mantuvo en el mismo nivel hasta 24 horas despu\u00e9s de la estimulaci\u00f3n. Adem\u00e1s, la actividad de uni\u00f3n al ADN de NF-\u03baB se observ\u00f3 tan pronto como 0,5 horas despu\u00e9s del tratamiento y aument\u00f3 r\u00e1pidamente a su valor m\u00e1ximo 2 horas despu\u00e9s, y luego disminuy\u00f3. Cuando se estimul\u00f3 RAW 264.7 con ABHG en presencia de un anticuerpo anti-TNF-\u03b1, la expresi\u00f3n del ARNm de TNF-\u03b1 no se vio afectada y la iNOS se regul\u00f3 negativamente.<\/p>\n<p>Estos resultados indican que la producci\u00f3n de TNF-\u03b1 por RAW 264.7, estimulada con un polisac\u00e1rido inmunomodulador como ABHG, se increment\u00f3 mediante la regulaci\u00f3n al alza de NF-\u03baB a trav\u00e9s de TLR4, y el TNF-\u03b1 reci\u00e9n producido mejor\u00f3 la producci\u00f3n de NO a trav\u00e9s de una v\u00eda autocrina.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Mizuno, M., &amp; Kawakami, S.
