(2014).\u00a0<a href=\"https:\/\/academic.oup.com\/humrep\/article\/29\/8\/1677\/2913977\">Antifibrotic properties of epigallocatechin-3-gallate in endometriosis<\/a>.\u00a0<i>Human Reproduction<\/i>, 29(8), 1677-1687.<\/p>\n<p>PREGUNTA DE ESTUDIO: \u00bfEl tratamiento con epigalocatequina-3-galato (EGCG) es eficaz en el tratamiento de la fibrosis en la endometriosis?<\/p>\n<p>RESPUESTA RESUMIDA: El EGCG parece tener propiedades antifibr\u00f3ticas en la endometriosis.<\/p>\n<p>LO QUE YA SE CONOCE: Histol\u00f3gicamente, la endometriosis se caracteriza por tejido fibroso denso que rodea las gl\u00e1ndulas endometriales y el estroma. Sin embargo, solo unos pocos estudios hasta la fecha han evaluado nuevas terapias candidatas para la fibrosis asociada a la endometriosis.<\/p>\n<p>DISE\u00d1O DEL ESTUDIO, TAMA\u00d1O, DURACI\u00d3N: Para este estudio de laboratorio, se analizaron muestras de 55 pacientes (45 con y 10 sin endometriosis) en edad reproductiva con ciclos menstruales normales. Un total de 40 ratones desnudos recibieron fragmentos endometriales proliferativos de inyecci\u00f3n \u00fanica de un total de 10 muestras.<\/p>\n<p>PARTICIPANTES \/ MATERIALES, AJUSTE, M\u00c9TODOS: Los efectos in vitro de EGCG y N- acetil- L -ciste\u00edna sobre marcadores fibr\u00f3ticos (alfa-actina de m\u00fasculo liso, col\u00e1geno tipo I, factor de crecimiento del tejido conectivo y fibronectina) con y sin estimulaci\u00f3n del factor de crecimiento transformante (TGF) -\u03b21, as\u00ed como en proliferaci\u00f3n celular, migraci\u00f3n e invasi\u00f3n y contracci\u00f3n de gel de col\u00e1geno de c\u00e9lulas estromales endometri\u00f3ticas y endometri\u00f3ticas se evaluaron mediante PCR en tiempo real, inmunocitoqu\u00edmica, ensayos de proliferaci\u00f3n celular, ensayos de migraci\u00f3n e invasi\u00f3n in vitro y \/ o ensayos de contracci\u00f3n de gel de col\u00e1geno. El in vitroLos efectos de EGCG sobre la prote\u00edna quinasa activada por mit\u00f3genos (MAPK) y las v\u00edas de se\u00f1alizaci\u00f3n de Smad en las c\u00e9lulas del estroma endometrial y endometri\u00f3tico se evaluaron mediante Western Blot. Adem\u00e1s, los efectos del tratamiento con EGCG sobre implantes endometri\u00f3ticos se evaluaron en un modelo de xenoinjerto de endometriosis en ratones desnudos inmunodeficientes.<\/p>\n<p>PRINCIPALES RESULTADOS Y EL PAPEL DE LA OPORTUNIDAD: El tratamiento con EGCG inhibi\u00f3 significativamente la proliferaci\u00f3n celular, la migraci\u00f3n y la invasi\u00f3n de las c\u00e9lulas del estroma endometrial y endometri\u00f3tico de pacientes con endometriosis. Adem\u00e1s, el tratamiento con EGCG disminuy\u00f3 significativamente el aumento dependiente de TGF-\u03b21 en la expresi\u00f3n de ARNm de marcadores fibr\u00f3ticos en c\u00e9lulas endometri\u00f3ticas y estromales endometriales. La contracci\u00f3n de geles de col\u00e1geno mediada por c\u00e9lulas del estroma endometri\u00f3sico y endometrial se atenu\u00f3 significativamente a las 8, 12 y 24 h despu\u00e9s del tratamiento con EGCG. La epigalocatequina-3-galato tambi\u00e9n inhibi\u00f3 significativamente la activaci\u00f3n estimulada por TGF-\u03b21 de las v\u00edas de se\u00f1alizaci\u00f3n de MAPK y Smad en las c\u00e9lulas del estroma endometrial y endometri\u00f3tico. Los experimentos con animales mostraron que EGCG previno la progresi\u00f3n de la fibrosis en la endometriosis.<\/p>\n<p>LIMITACIONES, MOTIVOS DE PRECAUCI\u00d3N: El atractivo del 3-galato de epigalocatequina como candidato a f\u00e1rmaco se ha visto disminuido por su biodisponibilidad relativamente baja. Sin embargo, se han patentado numerosas alteraciones de la mol\u00e9cula de EGCG, ya sea para mejorar la integridad del compuesto nativo o para generar una mol\u00e9cula m\u00e1s estable pero igualmente eficaz.
