Se examinaron la patolog\u00eda hep\u00e1tica, los metabolitos del \u00e1cido f\u00f3lico y la colina y la expresi\u00f3n g\u00e9nica en las v\u00edas del \u00e1cido f\u00f3lico y los l\u00edpidos.<\/p>\n<p>RESULTADOS: Los pesos del h\u00edgado y el bazo fueron m\u00e1s altos y los perfiles hematol\u00f3gicos se alteraron en los ratones alimentados con FASD. La histolog\u00eda hep\u00e1tica revel\u00f3 c\u00e9lulas degenerativas inusualmente grandes en ratones FASD Mthfr +\/\u2212 , compatible con la enfermedad del h\u00edgado graso no alcoh\u00f3lico. El \u00e1cido f\u00f3lico alto inhibi\u00f3 la actividad de MTHFR in vitro y la prote\u00edna MTHFR se redujo en ratones alimentados con FASD. Las proporciones de 5-metiltetrahidrofolato, SAM y SAM \/ S-adenosilhomociste\u00edna fueron menores en los h\u00edgados FASD y Mthfr +\/\u2212 . Los metabolitos de la colina, incluida la fosfatidilcolina, se redujeron debido al genotipo y \/ o la dieta en un intento de restaurar la capacidad de metilaci\u00f3n a trav\u00e9s de la s\u00edntesis de SAM dependiente de colina \/ beta\u00edna. Los \u00faltimos cambios, que inclu\u00edan una prote\u00edna 1 de uni\u00f3n al elemento regulador de esteroles nucleares m\u00e1s alta, un ARN mensajero (ARNm) de Srepb2 m\u00e1s alto, un ARNm del receptor X de farnesoide ( Nr1h4 ) m\u00e1s bajo y un ARNm de Cyp7a1 m\u00e1s bajo , conducir\u00edan a una mayor lipog\u00e9nesis y un catabolismo reducido del colesterol en la bilis.<\/p>\n<p>CONCLUSIONES: Sugerimos que un alto consumo de \u00e1cido f\u00f3lico reduce los niveles de actividad y prote\u00edna MTHFR, creando una pseudo-deficiencia de MTHFR. Esta deficiencia da como resultado la degeneraci\u00f3n de los hepatocitos, lo que sugiere un mecanismo de 2 golpes por el cual los hepatocitos mutantes no pueden adaptarse a las alteraciones de los l\u00edpidos y la integridad de la membrana alterada que surgen de los cambios en el metabolismo de los fosfol\u00edpidos \/ l\u00edpidos.
