Para dilucidar el mecanismo molecular que lleva a cabo su acci\u00f3n catab\u00f3lica sobre el hueso, se examin\u00f3 el efecto de la 1\u03b1, 25 dihidroxivitamina D3 (1\u03b1, 25 (OH) 2D3) sobre la osteoclastog\u00e9nesis y la expresi\u00f3n del ARNm de RANKL mediante cocultivo, RT \u2010 PCR y estudios de ejecuci\u00f3n nuclear..<\/p>\n<p>Al acelerar la tasa de transcripci\u00f3n del gen RANKL en las c\u00e9lulas osteobl\u00e1sticas SaOS2, 1\u03b1, 25 (OH) 2D3 mejor\u00f3 la formaci\u00f3n de osteoclastos in vitro a partir de monocitos perif\u00e9ricos. La clonaci\u00f3n y caracterizaci\u00f3n de la regi\u00f3n flanqueante 5 &#8216;del gen RANKL humano revel\u00f3 que el promotor b\u00e1sico comprende cajas TATA y CAAT invertidas flanqueadas por sitios de uni\u00f3n RUNX2.
