No se han reportado preocupaciones con respecto a su tolerancia y seguridad en humanos. Un estudio formal de fase I de seguridad, tolerancia y farmacocin\u00e9tica pareci\u00f3 justificado porque estos brotes se est\u00e1n utilizando como veh\u00edculos para la liberaci\u00f3n del glucosinolato glucorafanina y su isotiocianato af\u00edn sulforafano [1-isotiocianato- (4R) &#8211; (metilsulfinil) butano] en la ensayos.<\/p>\n<p>Estos ensayos se han dise\u00f1ado para evaluar la eficacia protectora contra el desarrollo de enfermedades neopl\u00e1sicas y otras. Se realiz\u00f3 un estudio cl\u00ednico aleatorizado, doble ciego y controlado con placebo de extractos de brotes que conten\u00edan glucosinolatos (principalmente glucorafanina, el precursor del sulforafano) o isotiocianatos (principalmente sulforafano) en voluntarios sanos que estaban hospitalizados en nuestra unidad de investigaci\u00f3n cl\u00ednica.<\/p>\n<p>Los sujetos se estudiaron en tres cohortes, cada una de las cuales inclu\u00eda tres individuos tratados y un receptor de placebo. Despu\u00e9s de un per\u00edodo de aclimataci\u00f3n de 5 d\u00edas con una dieta libre de cruc\u00edferas, los extractos de brotes de br\u00f3coli se administraron por v\u00eda oral a intervalos de 8 h durante 7 d\u00edas (21 dosis), y los sujetos fueron monitoreados durante este per\u00edodo y durante 3 d\u00edas despu\u00e9s del \u00faltimo tratamiento. . Las dosis fueron 25 micromol de glucosinolato (cohorte A), 100 micromol de glucosinolato (cohorte B) o 25 micromol de isotiocianato (cohorte C).<\/p>\n<p>La excreci\u00f3n acumulada media de ditiocarbamatos como fracci\u00f3n de la dosis fue muy similar en las cohortes A y B (17,8 +\/- 8,6% y 19,6 +\/- 11,7% de la dosis, respectivamente) y mucho mayor y m\u00e1s constante en la cohorte C (70,6%). +\/- 2,0% de la dosis). Se realizaron treinta y dos tipos de pruebas de hematolog\u00eda o qu\u00edmica antes, durante y despu\u00e9s del per\u00edodo de tratamiento.
