(2000). <a href=\"https:\/\/journals.physiology.org\/doi\/full\/10.1152\/ajpendo.2000.278.1.e146\">Glucose-induced changes in protein kinase C and nitric oxide are prevented by vitamin E<\/a>.\u00a0<i>American Journal of Physiology-Endocrinology and Metabolism<\/i>, 278(1), E146-E152.<\/p>\n<p>FONDO: Los cambios en la actividad o expresi\u00f3n de la prote\u00edna quinasa C (PKC), los productos reactivos del ox\u00edgeno y el \u00f3xido n\u00edtrico (NO) pueden explicar la alteraci\u00f3n del comportamiento celular que se observa en la diabetes. Se ha propuesto que estos cambios forman parte de la fisiopatolog\u00eda de la disfunci\u00f3n er\u00e9ctil.<\/p>\n<p>OBJETIVO: Buscamos determinar si las c\u00e9lulas del m\u00fasculo liso vascular del cuerpo cavernoso (CCSMC) cultivadas en un medio con alto contenido de glucosa exhiben cambios en la actividad y expresi\u00f3n de las isoformas de PKC, NO y productos reactivos de ox\u00edgeno, y averiguar si estos cambios son prevenidos por el \u03b1-tocoferol.<\/p>\n<p>METODOLOGIA: Se cultivaron CCSMC de rata en concentraciones de glucosa de 5, 15 y 30 mM durante 3, 7 y 14 d\u00edas. La expresi\u00f3n de isoformas de PKC se ensay\u00f3 con anticuerpos espec\u00edficos de isoformas. En los CCSMC cultivados en glucosa 30 mM durante 2 semanas, la isoforma PKC-\u03b22 se increment\u00f3 (n = 4; P &lt;0.01), mientras que la expresi\u00f3n de las isoformas \u03b1, \u03b4, \u03b5 y \u03b21 no se modific\u00f3. El NO, medido por la proporci\u00f3n de nitrato a nitrito, disminuy\u00f3 mucho a los 14 d\u00edas en 30 mM (n = 4; P &lt;0,002) en comparaci\u00f3n con glucosa 5 mM. Los productos de ox\u00edgeno reactivo se regularon al alza a los 14 d\u00edas cuando fueron analizados por la sonda fluorescente diclorofluoresce\u00edna diacetato bis (acetoximetil) (DCFH-DA) (n = 5; P &lt;0,01).<\/p>\n<p>RESULTADOS: Cuando estas mismas c\u00e9lulas se expusieron a \u03b1-tocoferol durante 14 d\u00edas, hubo una reducci\u00f3n de PKC-\u03b22 (57,8%; P &lt;0,01; n = 4) y una reducci\u00f3n en la formaci\u00f3n de productos de ox\u00edgeno reactivo (71,1%; P &lt;0,001; n = 4), junto con un aumento en la proporci\u00f3n de nitrato a nitrito (43,9%; P &lt;0,01, n = 4).<\/p>\n<p>CONCLUSION: Estos resultados sugieren que puede haber una interrelaci\u00f3n entre PKC, NO y la formaci\u00f3n de productos de ox\u00edgeno reactivo en CCSMC expuestos a un entorno de alta glucosa.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Ganz, M.
