(2020).\u00a0<a href=\"https:\/\/cyprusjmedsci.com\/en\/genistein-induced-apoptosis-affects-human-telomerase-reverse-transcriptase-activity-in-acute-promyelocytic-leukemia-131889\">Genistein-Induced Apoptosis Affects Human Telomerase Reverse Transcriptase Activity in Acute Promyelocytic Leukemia<\/a>.\u00a0<i>Cyprus J Med Sci<\/i>; 5(2): 153-6<\/p>\n<p>ANTECEDENTES \/ OBJETIVOS: El prop\u00f3sito de este estudio fue investigar los efectos de la geniste\u00edna sobre la actividad de la telomerasa y la apoptosis en una l\u00ednea celular de leucemia promieloc\u00edtica aguda (HL-60).<\/p>\n<p>MATERIAL y M\u00c9TODOS: En c\u00e9lulas HL-60, se evalu\u00f3 el efecto citot\u00f3xico de la geniste\u00edna disponible comercialmente mediante el m\u00e9todo XTT. El m\u00e9todo XTT es un ensayo de proliferaci\u00f3n celular. Se utiliz\u00f3 el m\u00e9todo de anexina V-EGFP para determinar la apoptosis. La transcriptasa inversa de la telomerasa humana (hTERT) es un marcador de la actividad de la telomerasa. El an\u00e1lisis de expresi\u00f3n del gen hTERT se realiz\u00f3 mediante RT-PCR en tiempo real LightCycler.<\/p>\n<p>RESULTADOS: En c\u00e9lulas HL-60, se encontr\u00f3 que la CI50 de geniste\u00edna era 50 \u00b5M a las 72 horas. Se observ\u00f3 que la inducci\u00f3n de apoptosis fue 4,25 veces mayor en comparaci\u00f3n con las c\u00e9lulas no tratadas con geniste\u00edna utilizadas como grupo de control. En comparaci\u00f3n con el grupo de control, se encontr\u00f3 que la actividad de hTERT disminuy\u00f3 en 5,16, 3,81 y 5,04 veces a las 24, 48 y 72 horas, respectivamente.<\/p>\n<p>CONCLUSI\u00d3N: La apoptosis inducida de c\u00e9lulas HL-60 por la reducci\u00f3n de la actividad de la transcriptasa inversa de la telomerasa humana puede ser un par\u00e1metro beneficioso en pacientes con leucemia.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Okcano\u011flu, T.
