Sin embargo, el mecanismo molecular de los efectos antiproliferativos de la geniste\u00edna no se comprende del todo. Usando ensayos de actividad promotora de transcriptasa inversa-PCR y hTERT, mostramos que la geniste\u00edna disminuy\u00f3 la expresi\u00f3n de hTERT y la actividad transcripcional de forma dosis-dependiente.<\/p>\n<p>Utilizando varias construcciones del promotor hTERT suprimidas, definimos que el promotor central de hTERT es suficiente para observar la represi\u00f3n inducida por geniste\u00edna de la actividad transcripcional de hTERT. Debido a que c-Myc participa en la regulaci\u00f3n transcripcional de hTERT, se examin\u00f3 la expresi\u00f3n de c-Myc. Se observ\u00f3 una disminuci\u00f3n dependiente de la dosis en el mensaje de c-Myc y las prote\u00ednas con el tratamiento con geniste\u00edna.<\/p>\n<p>Estos resultados indican que la geniste\u00edna reprime la actividad transcripcional de hTERT mediante la regulaci\u00f3n por disminuci\u00f3n de la expresi\u00f3n de c-Myc. Sin embargo, la represi\u00f3n de la actividad transcripcional de hTERT inducida por geniste\u00edna no fue bloqueada por la mutaci\u00f3n de c-Myc en el promotor de hTERT, lo que sugiere que hay factores adicionales implicados en la represi\u00f3n de la actividad de la telomerasa dependiente de geniste\u00edna.
