Aunque sus interacciones son necesarias para el plegado adecuado de hTERT naciente, as\u00ed como el ensamblaje de telomerasa activa, el papel preciso de las prote\u00ednas chaperonas en la regulaci\u00f3n de la localizaci\u00f3n nuclear de hTERT sigue sin estar claro.<\/p>\n<p>Aqu\u00ed demostramos que la curcumina inhibe la actividad de la telomerasa de una manera dependiente del tiempo y de la dosis al disminuir el nivel de expresi\u00f3n de hTERT. Despu\u00e9s del tratamiento con curcumina, observamos una clara acumulaci\u00f3n de hTERT en el compartimento citoplasm\u00e1tico de la c\u00e9lula. La retenci\u00f3n citoplasm\u00e1tica de hTERT inducida por la curcumina podr\u00eda deberse a una falla en la importaci\u00f3n nuclear, y la prote\u00edna hTERT citopl\u00e1smica resultante fue ubiquitinada y degradada r\u00e1pidamente por el proteasoma.<\/p>\n<p>Tambi\u00e9n informamos que el tratamiento con curcumina da como resultado una disminuci\u00f3n sustancial en la asociaci\u00f3n de p23 y hTERT, pero no afecta la uni\u00f3n de Hsp90 a hTERT. Por el contrario, el tratamiento del inhibidor de Hsp90 geldanamicina promueve la disociaci\u00f3n de las prote\u00ednas Hsp90 y p23 de hTERT.<\/p>\n<p>Tomados en conjunto, estos resultados demuestran que la interacci\u00f3n del complejo Hsp90-p23 con hTERT es fundamental para la regulaci\u00f3n de la localizaci\u00f3n nuclear de la telomerasa, y que la regulaci\u00f3n negativa de hTERT por la curcumina implica disociar la uni\u00f3n de hTERT con p23.<\/p>\n<p>Por lo tanto, la inhibici\u00f3n de la translocaci\u00f3n nuclear de hTERT por la curcumina puede proporcionar nuevas perspectivas para la regulaci\u00f3n de la actividad de la telomerasa durante la progresi\u00f3n tumorig\u00e9nica.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Lee, J.
