(2005). <a href=\"https:\/\/ar.iiarjournals.org\/content\/anticanres\/25\/5\/3293.full.pdf\">Expression profiles of apoptotic genes induced by curcumin in human breast cancer and mammary epithelial cell lines<\/a>.\u00a0<i>Anticancer research<\/i>, 25(5), 3293-3302.<\/p>\n<p>La curcumina (diferuloil metano), el pigmento diet\u00e9tico de color amarillo de los rizomas de la c\u00farcuma, ha sido reconocido como un agente quimiopreventivo debido a sus efectos antitumorales, antioxidantes y antiproliferativos.<\/p>\n<p>Los efectos citot\u00f3xicos, apopt\u00f3ticos y reguladores de genes tanto de la c\u00farcuma como de la curcumina se investigaron en la l\u00ednea celular de carcinoma de c\u00e1ncer de mama humano MCF-7 y se compararon con los efectos en c\u00e9lulas epiteliales mamarias humanas MCF-10A.<\/p>\n<p>Las c\u00e9lulas MCF-7 eran m\u00e1s sensibles a la c\u00farcuma y la curcumina que las c\u00e9lulas MCF-10A. Las c\u00e9lulas MCF-10A retuvieron comparativamente menos curcumina en el medio que las c\u00e9lulas MCF-7 despu\u00e9s de 24 h, reduciendo as\u00ed el efecto citot\u00f3xico.<\/p>\n<p>La curcumina indujo un porcentaje significativamente mayor de apoptosis en las c\u00e9lulas MCF-7 que en las c\u00e9lulas MCF-10A en todas las dosis. Se realiz\u00f3 la hibridaci\u00f3n de micromatrices de matrices apopt\u00f3ticas Clonetech con sondas de primera hebra marcadas de ARN total para identificar y caracterizar los genes regulados por la curcumina en las c\u00e9lulas tumorales. De los 214 genes asociados a la apoptosis en la matriz, la expresi\u00f3n de 104 genes fue alterada por el tratamiento con curcumina. La expresi\u00f3n g\u00e9nica se alter\u00f3 hasta niveles de 14 veces en MCF-7 en comparaci\u00f3n con solo hasta 1,5 veces en la l\u00ednea celular MCF-10A por curcumina.<\/p>\n<p>La curcumina regulaba positivamente (&gt; 3 veces) 22 genes y regulaba negativamente 17 genes (&lt;3 veces) a dosis de 25 microg \/ ml y 50 microg \/ ml en la l\u00ednea celular MCF-7. Los genes regulados al alza incluyen HIAP1, CRAF1, TRAF6, CASP1, CASP2, CASP3, CASP4, HPRT, GADD45, MCL-1, NIP1, BCL2L2, TRAP3, GSTP1, DAXX, PIG11, UBC, PIG3, PCNA, CDC10, JNK1 y RBP2. Los genes regulados negativamente fueron TRAIL, TNFR, AP13, IGFBP3, SARP3, PKB, IGFBP, CASP7, CASP9, TNFSF6, TRICK2A, CAS, TRAIL-R2, RATS1, hTRIP, TNFb y TNFRSF5. Si bien se observ\u00f3 un cambio en la expresi\u00f3n g\u00e9nica dependiente de la dosis en algunos genes, diferentes dosis de curcumina indujeron efectos reguladores opuestos en tres genes apopt\u00f3ticos.<\/p>\n<p>CONCLUSIONES: Estos resultados sugieren que la curcumina induce la apoptosis en las c\u00e9lulas del c\u00e1ncer de mama mediante la regulaci\u00f3n de m\u00faltiples v\u00edas de se\u00f1alizaci\u00f3n, lo que indica su uso potencial para la prevenci\u00f3n y el tratamiento del c\u00e1ncer.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Ramachandran, C., Rodriguez, S., Ramachandran, R., Nair, P.
