(2003). <a href=\"https:\/\/cancerres.aacrjournals.org\/content\/63\/20\/6825.full\">Induction of apoptosis by the garlic-derived compound S-allylmercaptocysteine (SAMC) is associated with microtubule depolymerization and c-Jun NH2-terminal kinase 1 activation.<\/a>\u00a0<i>Cancer research,<\/i>\u00a063(20), 6825-6837.<\/p>\n<p>CONTEXTO: Los estudios epidemiol\u00f3gicos y experimentales de carcinog\u00e9nesis proporcionan evidencia de que los componentes del ajo (Allium sativum) tienen actividad anticancer\u00edgena. Recientemente informamos que el derivado del ajo S-alilmercaptociste\u00edna (SAMC) inhibe el crecimiento celular en G2-M e induce apoptosis en c\u00e9lulas de c\u00e1ncer de colon humano (Shirin et al., Cancer Res., 61: 725-731, 2001).<\/p>\n<p>OBJETIVO: Debido a que una fracci\u00f3n de las c\u00e9lulas tratadas con SAMC se detienen espec\u00edficamente en la mitosis, examinamos el mecanismo de este efecto en el presente estudio.<\/p>\n<p>METODO Y RESULTADOS: La microscop\u00eda inmunofluorescente revel\u00f3 que el tratamiento de c\u00e9lulas SW480 o fibroblastos NIH3T3 con 150 \u03bcm de SAMC (la concentraci\u00f3n IC50) caus\u00f3 una r\u00e1pida despolimerizaci\u00f3n de microt\u00fabulos (MT), disrupci\u00f3n del citoesqueleto MT, fragmentaci\u00f3n del centrosoma y dispersi\u00f3n de Golgi en c\u00e9lulas en interfase. Tambi\u00e9n indujo la formaci\u00f3n de husos monopolares y multipolares en las c\u00e9lulas mit\u00f3ticas. Los ensayos de turbidez in vitro indicaron que SAMC actu\u00f3 directamente sobre la tubulina para provocar la despolimerizaci\u00f3n de la MT, aparentemente porque interact\u00faa con los grupos -SH de la tubulina. Para investigar las v\u00edas de se\u00f1alizaci\u00f3n implicadas en la apoptosis inducida por SAMC, analizamos la actividad quinasa c-Jun NH2-terminal (JNK) y descubrimos que el tratamiento con SAMC provoc\u00f3 una inducci\u00f3n r\u00e1pida y sostenida de la actividad de JNK. El inhibidor selectivo de JNK SP600125 inhibi\u00f3 la fase temprana (24 h) pero no la fase tard\u00eda (48 hy m\u00e1s tarde) de la apoptosis inducida por SAMC. La expresi\u00f3n de un mutante dominante negativo de JNK1 en c\u00e9lulas SW480 inhibi\u00f3 la apoptosis inducida por SAMC a las 24 h, pero no tuvo efecto protector a las 48 h. Los fibroblastos embrionarios de rat\u00f3n JNK1 &#8211; \/ &#8211; fueron resistentes a la apoptosis inducida por SAMC a las 24 h, pero no a las 48 h. Por otro lado, la inhibici\u00f3n o anulaci\u00f3n de la actividad de JNK1 no inhibi\u00f3 la detenci\u00f3n de G2-M inducida por SAMC. SAMC tambi\u00e9n activ\u00f3 la caspasa-3. El inhibidor de caspasa general z-VAD-fmk inhibi\u00f3 las fases temprana y tard\u00eda de la apoptosis inducida por SAMC.<\/p>\n<p>CONCLUSION: Concluimos que el compuesto derivado del ajo SAMC ejerce efectos antiproliferativos al unirse directamente a la tubulina e interrumpir el ensamblaje de MT, deteniendo as\u00ed las c\u00e9lulas en la mitosis y desencadenando las v\u00edas de se\u00f1alizaci\u00f3n de JNK1 y caspasa-3 que conducen a la apoptosis.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Xiao, D., Pinto, J.
