La HPB es causada por el crecimiento no maligno de elementos epiteliales y estromales de la pr\u00f3stata. La acci\u00f3n de IGF es importante para el crecimiento y desarrollo de la pr\u00f3stata, y se han documentado cambios en el sistema de IGF en los tejidos de BPH.<\/p>\n<p>Las principales v\u00edas de se\u00f1alizaci\u00f3n activadas por la uni\u00f3n de IGF-I al receptor de IGF-I (IGF-IR) son el brazo ERK de la cascada MAPK y la fosfoinositol-3-quinasa (PI3K) \/ prote\u00edna quinasa B (PKB \/ Akt) cascada.<\/p>\n<p>Probamos la hip\u00f3tesis de que SPE suprime el crecimiento e induce apoptosis en la l\u00ednea celular epitelial de pr\u00f3stata P69 al inhibir la se\u00f1alizaci\u00f3n de IGF-I. El tratamiento con 150 \u03bcg \/ ml SPE durante 24 h disminuy\u00f3 la proliferaci\u00f3n inducida por IGF-I de c\u00e9lulas P69 y la escisi\u00f3n inducida de la enzima poli (ADP-ribosa) polimerasa (PARP), Un \u00edndice de apoptosis.<\/p>\n<p>El tratamiento de c\u00e9lulas P69 privadas de suero con SPE de 150 \u03bcg \/ ml durante 6 h redujo la fosforilaci\u00f3n de Akt inducida por IGF-I (evaluada por transferencia Western) y la actividad de Akt (evaluada mediante un ensayo de quinasa Akt).<\/p>\n<p>El an\u00e1lisis de transferencia Western mostr\u00f3 que SPE redujo la fosforilaci\u00f3n inducida por IGF-I del sustrato receptor de insulina de la prote\u00edna adaptadora-1 y disminuy\u00f3 los efectos posteriores de la activaci\u00f3n de Akt, incluido el aumento de los niveles de ciclina D1 y la fosforilaci\u00f3n de la gluc\u00f3geno sintasa quinasa-3 y p70s6k .
